This study was compared the effect of shelf-life and DHA accumulation in chicken meat from broilers fed experimental diets for two weeks($21{\sim}35$ days) of growers. Two hundred-ten male Ross broilers, 1 day of age, were randomly allocated to seven treatment groups. Experimental diets were assigned to each of the seven groups: control diet containing tallow, T1 with 1.00% fish oil, T2 with 2.00% fish oil, T3 with 2.00% fish oil, 200 ppm vitamin E and 200 ppm vitamin C, T4 with 2.00% fish oil and 200 ppm vitamin C, T5 with 2.00% fish oil and 200 ppm vitamin E, and T6 with 3.00% fish oil. The levels of DHA in chicken meat was the highest in T6, and T1 in breast muscle and thigh muscle with skin was higher than that of $T2{\sim}T5,\;T2{\sim}T5$ in raw chicken meat and wing with skin was about two-fold higher than that of T1. The contents of DHA in chicken meat according to storage days were significantly reduced to 42.30%, 49.38% and 48.51% in T1, T2 and T6, respectively, and this decrease was higher than that of T3, T4 and T5 (p<0.05). Particularly, the rate of reduction of DHA was the lowest in the T3 and T5, which were the lowest in TBARS(thiobarbituric acid reactive substances). TBARS increased in the order of T6, T2 and T1, but reduced in the order of T3, T5 and T4 according to storage days, and there was a significant difference among the treatment groups (p<0.05).
The objectives of this study were to investigate the effects of dietary fish oil inclusion on the eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) contents and organoleptic characteristics of breast meat in mule ducks. Three hundred mule ducks at four weeks of age were randomly assigned to three dietary treatments with five replicate pens in each. One replicate pen had ten males and females each with a total of 100 ducks in each treatment. The diet in the three treatments contained 0, 1.5, and 3.0% fish oil, respectively. Body weights at 4, 6, 8, and 10 weeks of age, and feed efficiency at 4 to 6, 6 to 8, and 8 to 10 weeks of age were recorded. At 10 weeks of age, one male and one female from each replicate were sacrificed for oxidative stability of breast meat and the sacrificed males were employed for the analysis of fatty acids in breast meat and skin. Sensory evaluation of breast meat was also performed. A level of 3.0% fish oil in the diet significantly deteriorated feed efficiency and body weight gain. Dietary fish oil inclusion had a trend of increasing abdominal fat deposition and decreasing the flavor of breast meat. The EPA and DHA contents in the breast meat were higher than those in the breast skin irrespective of oil sources. The EPA and DHA contents in breast meat and breast skin was significantly increased in the 3.0% fish oil group. Although EPA and DHA were not efficiently deposited in the duck meat through dietary fish oil inclusion, this method can still provide a partial supplementation of EPA and DHA.
The relationship between microbial activity and physico-chemical characteristics of golf course soils were investigated in Kyonggi Province with different open years. The increasing tendency of soil PH with depth seemed to the result of mineral leaching. Exchangeable $Ca^2$+ , $Mg^2$+ and K+ showed relatively higher concentration in lower horizon indicating the leaching of minerals. The minerals were heavily leached in older golf course. There was a significant positive correlation between pH and $Ca^2$+.The contents of organic matters, total nitrogen and phosphorus were high in higher horizon.Dehydrogenase activity(DHA) and the content of ATP were determined as an index of soil microbial activity. DHA value was significantly different with golf club as well as golf courses ac-cording todifferent open year. This indicates that DHA was affected by several fertilizertreatments rather than herbicide pesticide treatments. DHA was positively correlated with water holding capacity and total nitrogen. The content of ATP was high in fairway and in golf course with older open year, showing the similar tendency with the results of DHA.
The relationships between microbial activity and disturbance level of soil were investigated from 15 parks in Seoul and undisturbed area. The physico-chemical characteristics of soil and dehydrogenase activity(DHA) as an index of soil microbial activity were analysed. There were ranges of 3.84~7.37 in pH, 9.63~40.33% in moisture content, 3.41~21.49% in organic matter, 0.36~0.79g/g in water holding capacity and 0.03~0.53% in total nitrogen investigated sites. DHA values of soil were 8.64~$146.76{\mu}g/g$ in park soil and 545.14~$1, 198.80{\mu}g/g$ in undisurbed area. DHA of park soil with high traffic density and contamination source from human activities was much lower than that of undisturbed area. DHA was positively correlated with moisture content, organic matter, water holding capacity and total nitrogen.
Glycerol dehydrogenases (GlyDHs) are essential for glycerol metabolism in vivo, catalyzing its reversible reduction to 1,3-dihydroxypropranone (DHA). The gldA gene encoding a putative GlyDH was cloned from Thermoanaerobacterium thermosaccharolyticum DSM 571 (TtGlyDH) and expressed in Escherichia coli. The presence of Mn2+ enhanced its enzymatic activity by 79.5%. Three highly conserved residues (Asp171, His254, and His271) in TtGlyDH were associated with metal ion binding. Based on an investigation of glycerol oxidation and DHA reduction, TtGlyDH showed maximum activity towards glycerol at 60℃ and pH 8.0 and towards DHA at 60℃ and pH 6.0. DHA reduction was the dominant reaction, with a lower Km(DHA) of 1.08 ± 0.13 mM and Vmax of 0.0053 ± 0.0001 mM/s, compared with glycerol oxidation, with a Km(glycerol) of 30.29 ± 3.42 mM and Vmax of 0.042 ± 0.002 mM/s. TtGlyDH had an apparent activation energy of 312.94 kJ/mol. The recombinant TtGlyDH was thermostable, maintaining 65% of its activity after a 2-h incubation at 60℃. Molecular modeling and site-directed mutagenesis analyses demonstrated that TtGlyDH had an atypical dinucleotide binding motif (GGG motif) and a basic residue Arg43, both related to dinucleotide binding.

Vitamin C ascorbic acid (AA) and dehydroascorbic acid (DHA) as an antioxidant have been shown to have protective effects in experimental neurological disorder models such as stroke, ischemia, and epileptic seizures. The present study was conducted to examine the protective effect of AA and DHA on Kainic acid (KA) neurotoxicity using organotypic hippocampal slice cultures (OHSC). After 12h KA treatment, significant delayed neuronal death was detected in CA3 region, but not in CA1. Intermediate dose of AA and DHA pretreatment significantly prevented cell death and inhibit ROS level, mitochondrial dysfunction and capase-3 activation in CA3 region. In the case of low or high dose, however, AA or DHA pretreatment were not effective. These data suggest that both AA and DHA pretreatment have neuroprotective effects on KA-induced neuronal injury depending on the concentration, by means of inhibition of ROS generation, mitochondrial dysfunction, and caspase-dependent apoptotic pathway.
The effects of eicosapentaenoic (EPA, 20:5n-3) and docosahexaenoic acids (DHA, 22:6n-3) on the phospholipase $A_2$ ($PLA_2$)-mediated release of arachidonic acid (AA, 20:4n-6) were studied in kidney microsomes from rats fed diets containing sunflower oil (SO) or fish oil (FO) concentrate for 11 months. The amounts of AA released by the endogenous $PLA_2$ enzyme were significantly lower by 38% in the FO, compared to the SO-fed rats (23.2 nmol versus 60.7 nmol AA released/mg protein/h in the FO- and SO-treated groups, respectively). The FO-derived microsomes released less linoleic acid (LA, 18:2n-6) and adrenic acid (22:4n-6), but larger amounts of the n-3 fatty acids, including EPA, DHA, docosapentaenoic acid (DPA, 22:5n-3), and 20:4n-3 than the SO-derived microsomes. A similar replacement of the AA and adrenic acid with the n-3 fatty acids including EPA and DHA was also observed in the microsomal phospholipid fraction from the FO-fed rats relative to the SO-treated group. The results suggest that the $PLA_2$-mediated release of AA is reduced and that of EPA is increased in compensation for AA decline in kidney microsomes from FO-fed rats (0.7 nmol EPA/mg protein/h versus 22.7 nmol EPA/mg protein/h for the SO and FO-treated groups). Replacement of the n-6 with n-3 fatty acids may explain the reduced synthesis of the AA-derived prostaglandins and the concomitant rise in the EPA-derived prostaglandins observed in kidneys of FO-treated rats.

Docosahexaenoic acid (DHA), an omega-3-fatty acid, modulates multiple cellular functions. In this study, we addressed the effects of DHA on human umbilical vein endothelial cell calcium transient and endothelial nitric oxide synthase (eNOS) phosphorylation under control and adenosine triphosphate (ATP, $100{\mu}M$) stimulated conditions. Cells were treated for 48 h with DHA concentrations from 3 to $50{\mu}M$. Calcium transient was measured using the fluorescent dye Fura-2-AM and eNOS phosphorylation was addressed by western blot. DHA dose-dependently reduced the ATP stimulated $Ca^{2+}$-transient. This effect was preserved in the presence of BAPTA (10 and $20{\mu}M$) which chelated the intracellular calcium, but eliminated after withdrawal of extracellular calcium, application of 2-aminoethoxy-diphenylborane ($75{\mu}M$) to inhibit store-operated calcium channel or thapsigargin ($2{\mu}M$) to delete calcium store. In addition, DHA ($12{\mu}M$) increased ser1177/thr495 phosphorylation of eNOS under baseline conditions but had no significant effect on this ratio under conditions of ATP stimulation. In conclusion, DHA dose-dependently inhibited the ATP-induced calcium transient, probably via store-operated calcium channels. Furthermore, DHA changed eNOS phosphorylation suggesting activation of the enzyme. Hence, DHA may shift the regulation of eNOS away from a $Ca^{2+}$ activated mode to a preferentially controlled phosphorylation mode.

Hydroxy acid has been used to enhance anti-aging and skin moisturization by peeling effect on the skin stratum corneum, and thus it has been widely used in topical products and cosmetic products. Among them, the effect that appears most effectively in a short period of time has been reported to be effected by the pH of the cosmetic formulations. However, there are many difficulties in use due to irritation caused by pH and concerns about side effects. The purpose of this study was to evaluate the effect of applying cosmetics with (1) varying concentrations, (2) types and (3) pH of hydroxy acid on human skin. 22 healthy adults were stained with DHA (dihydroxyacetone) and DC (dansyl chloride) on the forearm, and the skin exfoliation effect was measured after application of the test products. (1) The application of GA (glycolic acid) increased the desquamation by concentration dependent. (2) the test product prepared with neutral pH showed no exfoliation effect. In contrast, SA (salicylic acid) showed a statistically significant exfoliation effect at both acidic pH and neutral pH. (3) The neutral pH SA showed excellent exfoliation effect on bot DHA and DC stained stratum corneum. These results suggest that it is possible to manufacture safe cosmetics without damaging the skin barrier, providing an opportunity to use cosmetics that are expected to exfoliate to people, whose skin is sensitive to pH.
The differential effects of various fatty acids such as n-3 and n-6 types or degrees of unsaturation on the CYP2E1 induction and the production of lipid peroxidation (LPO) were investigated. The CYP2E1-transduced human hepatoma HepG2 cells (E47) were cultured in RPMI 1640 media containing different concentrations of various fatty acids up to 48 h incubation compared to 04 cells and CYP2E1-null cells. Treated fatty acids were linoleic acid (LA:n-6, C18:2), arachidonic acid (AA:n-6, C20:4) and docosahexaenoic acid (DHA:n-3, C22:6). The cell survival rate was decreased corresponding to the degree of unsaturation (LA>AA $\cong$DHA) and to LPO production in E47 and 04 cells. The four or five unsaturation degree of fatty acids, AA and DHA, caused time- and dose-dependent cell death in E47 cells but not as much as in C34 (without CYP2E1), suggesting an important role of CYP2E1 in the DHA mediated damage. In the levels of lipid peroxides (LPO), AA also elevated LPO by 3- and 5- fold compared to DHA or LA treated E47 cells. However, AA did not increase LPO until 48 h incubation in C34 cells. In conclusion, the polyunsaturated fatty acids induced CYP2E1 induction might be changed by the elevated levels of lipid peroxide (LPO) and oxidative stress through the connection of CYP2E1 and degrees of unsaturated fatty acids.
