Purification and Characterization of a Thermostable ${\beta}-1$,3-1,4-Glucanase from Laetiporus sulphureus var. miniatus

  • A ${\beta}-1$,3-1,4-glucanase from the fungus Laetiporus sulphureus var. miniatus was purified as a single 26 kDa band by ammonium sulfate precipitation, HiTrap Q HP, and UNO Q ion-exchange chromatography, with a specific activity of 29 U/mg. The molecular mass of the native enzyme was 52 kDa as a dimer by gel filtration. ${\beta}-1$,3-1,4-Glucanase showed optimum activity at pH 4.0 and $75^{\circ}C$. The half-lives of the enzyme at $70^{\circ}C$ and $75^{\circ}C$ were 152 h and 22 h, respectively. The enzyme showed the highest activity for barley ${\beta}$-glucan as ${\beta}-1$,3-1,4-glucan among the tested polysaccharides and p-nitrophenyl-${\beta}$-D-glycosides with a $K_m$, of 0.67 mg/ml, a $k_{cat}$ of 13.5 $S^{-1}$ and a $k_{cat}/K_m$ of 20 mg/ml/s.

Effects of Vitamin E Supplementation on Renal Lipid Peroxidation in High Fat Diet and Adriamycin Induced Experimental Nephrotic Syndrome Model Rats (고지방식이와 Adriamycin으로 유도된 신증후군 흰쥐실험모델에 비타민 E 첨가식이가 신장의 지질과산화대사에 미치는 영향)

  • This study was conducted to investigate the effects of vitamin E supplementation renal lipid peroxidation in high fat diet and adriamycin (ADR) induced experimental nephrotic syndrome model rats. Treated rats were injected intraperitoneally with ADR (2mg/kgBW/wk) once a week for four weeks. control rats were injected with saline solution instead of ADR. The rats in each group were fed experimental diets of three levels of vitamin E for 10 weeks: Normal (501U/kg diet), high (5,000IU/kg diet), excess (7,500IU/kg diet). The high fat diet and ADR treatment was performed to induce the decrease of kidney functions. Serum total cholesterol was significantly decreased by the excess supplementation. But there was no effect of vitamin E supplementation on serum total lipid and triglyceride. Thiobarbituric acid reacting substances(TBARS) was significantly decreased at high and excess supplementation. Glutathione reductase (GR), glutathione peroxidase ({TEX}$GP_{x}${/TEX}) and catalase activities (CAT) were measured as antioxidative enzymes. The renalglutathione reductase (GR) and catalase activities (CAT) were inclined to elevate by vitamin E supplementation. Thus the vitamin E supplementation was found to have an antioxicant effect. These results suggested that vitamin E supplementation could alleviate the changes in renal lipid peroxidation.

  • Partially purified water-soluble extract mixture from Ricini and Coptidis (named as RIC) showed to be a potent inhibitor of human immunodeficiency virus-1 (HIV-1) replication. RIC was evaluated for in vitro anti-HIV activity using SupTl and H9 cells infected by a recombinant virus (pSVCAT) containing chloramphenicol acetyltransferase (CAT) gene substituted for nef gene in the HIV-1 genome. RIC inhibited syncytiaformation of SupTl cells with a half maximal effective concentration, $IC_{50}$/, of 2.5 $\mu\textrm{g}$/mι and showed marked inhibition of CAT activity in the infected H9 cells and also suppressed reverse transcriptase (RT) activity in the supernatant of the infected H9 culture. However, RIC did not inhibit the activity of reverse transcriptase directly when it was mixed with the enzyme or with viral particles. Berberine, one of components of RIC, also showed similar anti-HIV activity as RIC did. The data suggest that there are active ingredients which mediate anti-HIV activity in RIC.

  • Human immunodeficiency virus type 1 (HIV-1) contains several nonstructural genes which are required for the viral replication and disease pathogenesis. Among them, tat and nef genes encode an essential transactivator of HIV-1 LTR and a pluripotent protein which seems to be essential for the in vivo but not in vitro viral replication, respectively. We constructed two tat and n of defective HIV-1 and tested for their ability to replicate in several T cells. The defective viruses did not replicate in CD4$\^$+/ T cells, but rescued in the recombinant Jurkat-tat cell which also contains tat gene. The replication of tat and nef defective HIV-1 which expresses chloramphenicol acetyltransferase(CAT) gene was easily detected by a sensitive CAT assay. No revertant was identified during the passages of the mutant viruses for more than two months in Jurkat-tat cells. tat and n of defective HIV-1 could be used instead of wild type viruse for several purposes such as inhibitor screening and development of attenuated AIDS vaccine.

Inabenfide-Induced Alleviation of Salt Stress in Rice as Linked to Changes in Salicylic Acid Content and Catalase Activity

  • The effect of inabenfide was investigated in rice seedlings subjected to salt stress in relation to changes in chlorophyll fluorescence(${\Delta}F/Fm'$), lipid peroxidation, salicylic acid(SA) content, and catalase(CAT) activity. A reduction in shoot growth of rice seedlings by 120 mM NaCl treatment was significantly alleviated by pretreatment with 30 ${\mu}M$ inabenfide. Sodium ion content was not affected by pretreatment with inabenfide, suggesting that alleviation was not due to a reduction in sodium ion uptake by the rice seedlings. At three days after NaCl treatment, the rice seedlings pretreated with inabenfide showed a higher ${\Delta}F/Fm'$(30%) and lower lipid peroxidation(28%) compared with the rice seedlings treated with NaCl alone. After NaCl treatment, CAT activity in the third leaf of rice seedlings decreased significantly but alleviated by pretreatment with inabenfide. Furthermore, pretreatment with inabenfide also reduced the level of SA which accumulated drastically in the third leaf of rice seedlings within a day after exposure to salt stress. These results suggest that inabenfide prevents SA accumulation in rice seedlings under salt stress which eventually induces the alleviation of salt stress damage.

Changes in Hematological Responses and Antioxidative Enzyme Activities of Japanese Eel Anguilla japonica Exposed to Elevated Ambient Nitrite (아질산에 노출된 뱀장어(Anguilla japonica)의 혈액학적 반응과 항산화효소의 활성 변화)

  • The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

  • This study analyzes the content of YouTube channel "Cream Heroes" dealing pet(cat) produced in South Korea currently. Due to the advance of science and technology in the 21 century, the commons are able to independently upload their own content after filming videos, and it is called One Person Media. Unlike existing One Person Media that contains provocative and violent factors, "Cream Heroes" is sound content that shows various features of cats. Specifically, this study explores how and why "Cream Heroes" is popular via content analysis. Throughout this, it studies not only the trait of popular YouTube channel, but also proposes the proper environment of One Person Media.

Computed Tomography and Magnetic Resonance Imaging Features of Spinal Chondrosarcoma in a Cat

  • An 8-year-old, spayed female Persian cat weighing 3.6 kg presented with a lumbosacral mass and bilateral weight bearing hindlimb lameness. Computed tomography (CT) and magnetic resonance imaging (MRI) revealed a dumbbell-shaped heterogeneous mass extending through the internal surface of the ileum and surrounding the lumbosacral junction. CT also revealed extensive osteoproliferation and bone lysis of the sacrum, but no evidence of any pulmonary metastasis. Furthermore, MRI revealed a focal area in the spinal cord showing connection with the adjacent tumor, suggesting tumor invasion into the spinal cord. Low-grade myxoid chondrosarcoma was histopathologically diagnosed. This is the first report describing CT and MRI findings of spinal cord chondrosarcoma in veterinary medicine. This study suggests that combining CT with MRI is a more sensitive tool for evaluating spinal tumors than using CT or MRI alone.

  • Software Defect Prediction (SDP) enhances the efficiency of software development by proactively identifying modules likely to contain errors. A major challenge in SDP is improving prediction performance. Recent research has applied deep learning techniques to the field of SDP, with the SAINT model particularly gaining attention for its outstanding performance in analyzing structured data. This study compares the SAINT model with other leading models (XGBoost, Random Forest, CatBoost) and investigates the latest deep learning techniques applicable to SDP. SAINT consistently demonstrated superior performance, proving effective in improving defect prediction accuracy. These findings highlight the potential of the SAINT model to advance defect prediction methodologies in practical software development scenarios, and were achieved through a rigorous methodology including cross-validation, feature scaling, and comparative analysis.

  • As the children with Autistic Spectrum Disorders(ASD) are increasing, and study of intelligent robot is dynamically proceeding, therapeutic robots are needed to help ASD children who have a lack of social interaction, inadequacy of communication. This paper deals with the design and operation of a cat robot for the treatment. To configure the behavior based on the results of field experiments, emotional treatment is suggested and robot structure considering ASD characteristic is designed with some sensors. In addition, we verified the effectiveness of the operation by field test.
