Neuroprotective & antioxidant effects of diets high in n-6 and n-3 fatty acids in rat focal brain ischemia model (N-6와 n-3 지방산이 풍부한 식이가 뇌졸중 유발 모델에서 뇌경색 크기 및 항산화 효소계에 미치는 영향)

  • This study was undertaken to investigate the effects of n-6(corn oil) & n-3(fish oil) fatty acids on infarction size and the cerebral activities of antioxidant enzyme in rat focal brain ischemia model. Weaning Sprague-Dawley rats were fed with either corn oil supplemented diet(COD, 14% corn oil) or fish oil supplemented diet(FOD, 14% menhaden oil) for 6 weeks. The right middle cerebral artery was occluded for 2 hours with a silicon rubber coated nylon surgical thread. After 24 hours of recirculation, the rats were sacrificed and brain sections were photographed using CCD camera after staining with 2, 3, 5-triphenyltetrazolium chloride for 60 minutes in room temperature. The infarcted area was measured and the volume of infarction was calculated. Catalase(CAT), superoxide dismutase(SOD) activities, and fatty acid composition in the brain were also measured. The total and corrected infarction volumes were not significantly different between FOD and COD group. The docosagexaenoic acid(DHA) and DHA content/arachidonic acid(AA) ratio of the cerebral cortex, an index of defense against lipid oxidation, were significantly increased in FOD group compared to those of COD group(p<0.05). In the left cortex(non-infarction side) as well as the right cortex(infarction side) of FOD group, CAT and Cu/Zn SOD activities were higher than those of the COD group(p<0.05). However, CAT and Cu/Zn SOD activities were not significantly different between the left cortex(non-infarction side) and the right cortex(infarction side) of both FOD and COD group. GPx activities were also not significantly different between two groups. Our results demonstrate that the brain infarction size in FOD and COD were not significantly different. However, cerebral lipid composition and antioxidant enzyme activities in FOD and COD group were different. Fish oil, a source of n-3 polyunsaturated fatty acid(PUFA) and corn oil, that of n-6(PUFA) may have a protective effect against oxidative stress induced via different mechanisms.

Evaluation of Cat Brain infarction Model Using MicroPET (마이크로 PET을 이용한 고양이 뇌 경색 모델의 평가)

  • Purpose: PET has some disadvantage in the imaging of small animal due to poor resolution. With the advent of microPET scanner, it is possible to image small animals. However, the image quality was not good enough as human image. Due to larger brain, cat brain imaging was superior to mouse or rat. In this study, we established the cat brain infarction model and evaluate it and its temporal charge using microPET scanner. Materials and Methods: Two adult male cats were used. Anesthesia was done with xylazine and ketamine HCl. A burr hole was made at 1cm right lateral to the bregma. Collagenase type IV 10 ${\mu}l$ was injected using 30 G needle for 5 minutes to establish the infarction model. $^{18}F$-FDG microPET (Concorde Microsystems Inc., Knoxville, TN) scans were performed 1, 11 and 32 days after the infarction. In addition, $^{18}F$-FDG PET scans were performed using human PET scanner (Gemini, Philips medical systems, CA, USA) 13 and 47 days after the infarction. Results: Two cat brain infarction models were established. The glucose metabolism of an infarction lesion improved with time. An infarction lesion was also distinguishable in the human PET scan. Conclusion: We successfully established the cat brain infarction model and evaluated the infarcted lesion and its temporal change using $^{18}F$-FDG microPET scanner.

A Encryption Technique of JPEG2000 Image Using 3-Dimensional Chaotic Cat Map (3차원 카오스 캣맵을 이용한 JPEG2000 영상의 암호화 기술)

  • In this paper, we proposed the image hiding method which decreases calculation amount by encrypt partial data using discrete wavelet transform(DWT) and linear scale quantization which were adopted as the main technique for frequency transform in JPEG2000 standard. Also we used the chaotic system and cat map which has smaller calculation amount than other encryption algorithms and then dramatically decreased calculation amount. This method operates encryption process between quantization and entropy coding for preserving compression ratio of images and uses the subband selection method. Also, suggested encryption method to JPEG2000 progressive transmission. The experiments have been performed with the Proposed methods implemented in software for about 500 images. Consequently, we are sure that the proposed is efficient image encryption methods to acquire the high encryption effect with small amount of encryption. It has been shown that there exits a relation of trade-off between the execution time and the effect of the encryption. It means that the proposed methods can be selectively used according to the application areas.

  • Cytochrome P450 enzymes have been intensively investigated in hepatic tissues and several mammalian cell lines. Compared to most studies about cytochrome P450 isozymes in liver in vivo and hepatic, cell lines in vitro, the study of cytochrome P450IA1 in human breast cancer cells could be very important to understand the mechanism of the regulation of CYPIA1 gene expression and cell growth. MCF-7 human breast cancer cells are well characterized to study estrogen and antiestrogen action due to the fact that they contain high level of estrogen receptor and have biological markers characterized. And also MCF-7 cells express high level of arylhydrocarbon hydroxylase activity and human cytochrome P450IA1 cDNA was cloned from MCF-7 cells. Ah receptor was characterized in many breast cancer cell lines and polycyclic aromatic hydrocarbon such as 3-MC induced the expression of CYPIA1 gene and cytochrome P450- dependent monooxygenase activity. We undertook a study to examine the effect of estrogens and other chemicals on the regulation of human CYPIA1 gene expression in MCF-7 cells via RTPCR analysis, that might help us to understand the mechanism of the regulation of CYPIA1 gene expression and MCF-7 cell growth. Expression vector containing the functional 5'-regulatory region of human CYPIA1 fused to the CAT reporter gene was transfected into estrogen receptor positive MCF-T cells or estrogen receptor negative MDA-MB-231 cells. After these cells were treated with various chemicals, RTPCR was carried out to measure both CYPIA1 mRNA and CAT mRNA levels. 1nM 3-MC increased in both P450 and CAT mRNA levels over those of control by two folds in MCF-7 cells but does not in MDA-MB-231 cells. Estrogen or tamoxifen or retinoic acid or chrysin decreased in both P450 and CAT mRNA levels that were induced by 3-MC in MCF-7 when each chemical was administered with 3-MC concomitantly. These results suggested that the level of CYPIA1 gene expression is modulated with estrogen-related molecules and make it possible to speculate that ER is related to CYPIA1 gene expression and cell growth in breast cancer cells. Effect of Pyroligneous Liquor on Oxygen Radicals and Their Scavenger Enzymes in Liver of CD Rats (흰쥐 간조직의 활성산소 및 제거효소에 미치는 목초액의 영향)

  • This study was designed to investigate the effects of pyroligneous liquor on oxygen radicals and their scavenger enzymes in the liver of Cri/Bgi CD rats (7 rats per group). Male rats were fed a basic diet prepared in our Lab., PL-0 (Control), PL-1, PL-25, PL-50 and PL-75 groups were Prepared to be 0%, 1%, 25%, 50% and 75%with distilled water using pyroligneous liquor (35% of Choa Co. Ltd.), and were administrated orally for 8 weeks. Superoxide radical contents in liver mitochondria and microsomes were significantly decreased to 12-14%, 11-15%, respectively, in these PL-25 and PL-50groups compared with the control group. Hydroxyl radical content in mitochondria and microsomes were markedly decreased to 12-20% and 17%, respectively, in these PL-25 and PL-50% groups compared with the control group. Hydrogen peroxide content in mitochondria and microsomes were significantly decreased about 15-12% and 22-20% in liver of PL-25 and PL-50 groups compared with the control group. Mn-SOD and Cu/Zn-SOD activities in liver of PL-25 and PL-50 groups were remarkably increased to 15-25%, 11-16%, respectively, compared with the control group. GPx activities in mitochondria and microsomes were significantly increased in the liver of PL-25 and PL-50 groups compared with the control group. CAT activities in mitochondria and cytosol were significantly increased to 12-14%, 15-27%, respectively, in the liver of PL-25 and PL-50 groups compared with the control group. These results suggest that long term administration orally of 25 and 50% pyroligneous liquor may effectively inhibit the formation of oxygen free radicals, and also scavenger enzyme activities significantly increase through the administration orally.

Bandwidth Reservation and Call Admission Control Mechanisms for Efficient Support of Multimedia Traffic in Mobile Computing Environments (이동 컴퓨팅 환경에서 멀티미디어 트래픽의 효율적 지원을 위한 대역폭 예약 및 호 수락 제어 메커니즘)

  • One of the most important issues in guaranteeing the high degree of QoS on mobile computing is how to reduce hand-off drops caused by lack of available bandwidth in a new cell. Each cell can request bandwidth reservation to its adjacent cells for hand-off calls. This reserved bandwidth can be used only for hand-offs, not for new calls. It is also important to determine how much of bandwidth should be reserved for hand-off calls because reserving too much would increase the probability of a new call being blocked. Therefore, it is essential to develop a new mechanism to provide QoS guarantee on a mobile computing environment by reserving an appropriate amount of bandwidth and call admission control. In this paper. bandwidth reservation and call admission control mechanisms are proposed to guarantee a consistent QoS for multimedia traffics on a mobile computing environment. For an appropriate bandwidth reservation, we propose an adaptive bandwidth reservation mechanism based on an MPP and a 2-tier cell structure. The former is used to predict a next move of the client while the latter to apply our mechanism only to the client with a high hand-off probability. We also propose a call admission control that performs call admission test only on PNC(Predicted Next Cell) of a client and its current cell. In order to minimize a waste of bandwidth caused by an erroneous prediction of client's location, we utilize a common pool and QoS adaptation scheme. In order evaluate the performance of our call admission control mechanism, we measure the metrics such as the blocking probability of new calls, dropping probability of hand-off calls, and bandwidth utilization. The simulation results show that the performance of our mechanism is superior to that of the existing mechanisms such as NR-CAT2, FR-CAT2, and AR-CAT2.

A Case of Feline Proliferative Eosinophilic Keratitis Treated by Topical Cyclosporine and Corticosteroids (고양이 증식성 호산구성 각막염에서 국소적인 사이클로스포린과 코르티코스테로이드 점안 1례)

  • A 6-year-old spayed female Persian cat presented with a 3-month history of recurrent ulcerative keratitis with noticeable opacification and vascularization of the right cornea. The lesion was nonresponsive to topical antibiotics and to nonsteroidal anti-inflammatory drugs. Ophthalmic examination showed signs of ocular discomfort, such as epiphora and blepharospasm, in the right eye. Biomicroscopic examination revealed an irregular, edematous, vascularized mass with pink to white tissue on the entire cornea and mild conjunctivitis. A tentative diagnosis of feline proliferative eosinophilic keratitis (FPEK) was made on the basis of clinical appearance. Cytologic examination of the cornea showed a mixture of numerous eosinophils and mast cells, which confirmed the original diagnosis of FPEK. The cat was treated with a topical antibiotic-corticosteroid combination, cyclosporine ointment, trifluridine eye drops, and oral Llysine. The clinical signs improved remarkably 18 days after the cat was first examined. The short-term use of corticosteroids and long-term use of cyclosporine and an anti-viral agent resolved the lesion without recurrence of the disease for 1 year.

Correlation of Executive Function and Quantitative Electroencephalography in Children and Adolescents with Attention-Deficit/Hyperactivity Disorder (주의력결핍 과잉행동장애 소아청소년의 실행기능과 정량화 뇌파의 상관성 연구)

  • Objectives : Attention-deficit hyperactivity disorder(ADHD) is characterized by significant impairments in executive functions, with a prevalence of approximately 3-5% of all children worldwide. The goal of this study was to examine the relationship between executive functions and electrophysiological activities in children and adolescents with ADHD. Methods : In 31 patients with ADHD, resting-state EEG was recorded, and Comprehensive Attention Test(CAT), Stroop Color-Word Inference Test(Stroop CWIT), Trail Making Test(TMT), and Wisconsin Card Sorting Test(CST) were administered. Korean version of the ADHD Rating Scale(K-ARS) was assessed. Results : Alpha and beta power positively correlated with the Attention Quotient(AQ), while delta power negatively correlated with AQ from CAT. In the Stroop CWIT, decreased delta power and increased beta power were related to higher performance. Power of the alpha band increased with higher TMT performance. Moreover, delta power negatively correlated with good performance on the CST, while alpha and high gamma band showed a positive correlation. Correlation with the parent-rating of ADHD symptoms showed a negative correlation between alpha power and higher scores on the K-ARS. Conclusions : These findings indicate that relative power in higher frequency bands of EEG is related to the higher executive function in children and adolescents with ADHD, while the association with the relative power in lower frequency bands of EEG seem to be vice versa. Furthermore, the findings suggest that QEEG may be a useful adjunctive tool in assessing patients with ADHD.

The Anti-oxidative Activities of Fermented Hibiscus syriacus Extract in High Fat Diet-induced obese C57BL/6 Mice (고지방식이로 유도된 비만 C57BL/6 쥐에서 발효 무궁화 추출물의 항산화 활성)

  • The purpose of this study was to evaluate the anti-oxidative effects of fermented Hibiscus syriacus extract on obese mice. The antioxidant experiments were conducted both in vivo and in vitro. The in vitro experiment was measured through the use of nitric oxide (NO) free radical scavenging activity. The results showed that the scavenging activity of the NO free radical in fermented Hibiscus syriacus extract had higher activity than unfermented Hibiscus syriacus extract. The in vivo experiments were divided into a normal dietary group, a high fat dietary group (CON), a high fat dietary + fermented Hibiscus syriacus extract group (FH), and a high fat dietary + unfermented Hibiscus syriacus extract group. These four groups received intraperitoneal administrations for four weeks. Next, the levels of superoxide dismutase (SOD), catalase (CAT), and malondialdehyde (MDA) in a liver homogenate were measured. The results showed that the levels of SOD and CAT in FH were higher than the CON and MDA levels, which were significantly decreased in FH. Consequently, fermented Hibiscus syriacus extract had an inhibitive effect on oxidative stress and a heightening effect on antioxidant enzyme activity in obese mice, suggesting that fermented Hibiscus syriacus extract could be utilized as a useful, natural antioxidant.

Hemipelvectomy in a Cat with Obstipation (심한 변비를 보이는 고양이에서 반골반절제술을 이용한 외과적 치료 증례)

  • A 9-month-old female Korean short hair cat weighing 2.2 kg presented for evaluation of a two-week history of obstipation. The owner reported that the cat sustained pelvic fractures 4 months previous to the onset of fecal tenesmus. On physical examination, fecal tenesmus was observed and restriction of the movement of the right coxofemoral joint was evident. Rectal palpation revealed narrowing of the pelvic canal with a hard bony protuberance at the bilateral acetabulum and pubic bones. Radiographs revealed a distended colon with feces and narrowing of the pelvic canal with abnormal structure of the pelvic bone. Conservative management consisting of stool softeners and a warm water enema was instituted; however, there was no improvement in obstipation. Partial iliac, ischial, pubic, and acetabular ostectomies were performed. Postoperative radiographs and rectal palpation revealed the enlarged pelvic canal. Stool softeners (5 ml orally twice daily) was administered following surgery for 14 days and then tapered down to 2.5 ml for 14 days. A warm water enema was performed twice postoperatively. At examination 14 days postoperatively, no problems with defecation and gait were reported. There was no evidence of obstipation and lameness of the left pelvic limb 5 months postoperatively.
