  • In the cases of the experimentally destroyed receptor organ of the Corti of the cats, "electric cochlea" were inserted near the auditory neuron through the Scala tympani as an input of the inner device, and outer device is placed near the receiver of the audiometer. During exposing noise through the outer device, kymographic record were obtained as following: 1) Correlation between increasing intensity and amplitude showed parallel responses 2) The auricular reflex by repeated stimulation of the sound showed considerable increase at first, and decrease respectively. 3) In this experimental animals, absolutely non-responsed period, relatively non-responsed period and responsed period were observed. 4) Above mentioned reflex indicate that "Auditory sensation" can be induced by inserted "Electric cochlea" in the cases of the experimentally destroyed receptor organ of Corti of the cats.

Transcriptional Regulation of the Drosophila Proliferating Cell Nuclear Antigen Gene and raf Proto-oncogene by Ursolic Acid in Drosophila Cultured Kc Cells

  • Promoter of the Drosophila proliferating cell nuclear antigen (PCNA) gene contains DRE (Drosophila DNA replication-related element) required for the high level expression of replication-related genes. Recently, we found that promoter region of the D-raf (a Drosophila homolog of the human c-raf-1) contains two sequences homologous to the DRE and demonstrated the DRE/DREF (DRE-binding factor) involvement in regulation of the D-raf gene. In this study, using ursolic acid (UA), a pentacyclic triterpene acid reported to possess antitumor activities, we examined effects of UA on proliferation of the Drosophila cultured Kc cells and on expression of the PCNA and D-raf genes. UA showed an inhibitory effect on proliferation of the Kc cells in a concentration-dependent manner in DNA content assays and [3H]thymidine incorporation assays. The IC50 value of anti-proliferative effects of UA in DNA content assays was about 7.5uM. UA showed inhibitory effects on expression of the PCNA as well as on that of the D-raf, which were examined with the reporter plasmic p5'-168DPCNACAT or p5'-878DrafCAT, respectively. The results obtained in the present study suggest that expression of the PCNA and D-raf genes is coordinately regulated in at least UA-treated Kc cells and that down-regulation of expression of the PCNA and D-raf genes might be related with the antitumor activities of UA.

Excretion of Toxoplasma gondii oocysts from Feral Cats in Korea

  • Sporulated oocysts from the feces of infected cats with Toxoplasma gondii can cause detrimental disease in both humans and animals. To investigate the prevalence of feral cats that excrete T. gondii oocysts in the feces, we examined fecal samples of 563 feral cats over a 3-year period from 2009 to 2011. Oocysts of T. gondii excreted into the feces were found from 4 of 128 cats in 2009 (3.1%) and one of 228 (0.4%) in 2010 while none of the 207 cats in 2010 were found positive with oocysts in their feces, resulting in an overall prevalence rate of 0.89% (5/563) between 2009 and 2011. Among the 5 cats that tested positive with T. gondii oocysts, 4 of the cats were male and 1 was a female with an average body weight of 0.87 kg. Numerous tissue cysts of 60 ㎛ in diameter with thin (<0.5 ㎛) cyst walls were found in the brain of one of the 5 cats on necropsy 2 months after the identification of oocysts in the feces. A PCR amplification of the T. gondii-like oocysts in the feces of the positive cats using the primer pairs Tox-5/Tox-8 and Hham34F/Hham3R confirmed the presence of T. gondii oocysts in the feces. This study provides a good indication of the risk assessment of feral cats in the transmission of T. gondii to humans in Korea.

CHANGES IN INTRAPULPAL NERVE ACTIVITY AND OCCLUDING ASPECTS OF DENTINAL TUBULES BY DENTIN DESENSITIZER CONTAINING GLUTARALDEHYDE (Glutaraldehyde계 상아질 과민증 탈감작제에 의한 치수신경 활동성 변화 및 상아세관 폐쇄양상)

  • The effects of application of dentin desensitizer containing glutaraldehyde (Gluma Desensitizer) and dentin adhesive system (All Bond 2) to the exposed dentin on the intradental nerve activity (INA) and the occluding aspects of dentinal tubules were investigated in cat canine teeth. Single pulp nerve units were dissected from the inferior alveolar nerve and indentified as $A{\delta}$-fiber units. The INAs elicited by 4M NaCl before and after the application of each experimental agent were compared. The morphological changes of exposed dentin surfaces and dentinal tubules in the dentin specimens used to record INAs were observed by SEM. The results obtained were as follows. 1. Eight $A{\delta}$-fiber units (conduction velocity: $8.0{\pm}4.0m$/sec) were identified. 4M NaCl evoked an irregular burst of action potentials which ceased immediately after washing. 2. In 4 $A{\delta}$-fiber units, the change of INA after the application of Gluma Desensitizer was $133.9{\pm}80.7%$ when it was compared with the INA before the application of the same agent. 3. In 4 $A{\delta}$-fiber units, application of All Bond 2 completely abolished the INA induced by 4M NaCl. 4. In specimens applied with Gluma Desensitizer, the formation of hybrid layer as well as the identification of resin penetration and reaction products with proteins in dentinal tubules were not clearly observed in interface between dentin and adhesive resin. In specimens applied with All Bond 2, the gap width of 2-$3{\mu}m$ was formed between exposed dentin and adhesive resin, and the hybrid layer and resin tags were not clearly formed either.

Application of Machine Learning Techniques for Problematic Smartphone Use (스마트폰 과의존 판별을 위한 기계 학습 기법의 응용)

  • Purpose - The purpose of this study is to explore the possibility of predicting the degree of smartphone overdependence based on mobile phone usage patterns. Design/methodology/approach - In this study, a survey conducted by Korea Internet and Security Agency(KISA) called "problematic smartphone use survey" was analyzed. The survey consists of 180 questions, and data were collected from 29,712 participants. Based on the data on the smartphone usage pattern obtained through the questionnaire, the smartphone addiction level was predicted using machine learning techniques. k-NN, gradient boosting, XGBoost, CatBoost, AdaBoost and random forest algorithms were employed. Findings - First, while various factors together influence the smartphone overdependence level, the results show that all machine learning techniques perform well to predict the smartphone overdependence level. Especially, we focus on the features which can be obtained from the smartphone log data (without psychological factors). It means that our results can be a basis for diagnostic programs to detect problematic smartphone use. Second, the results show that information on users' age, marriage and smartphone usage patterns can be used as predictors to determine whether users are addicted to smartphones. Other demographic characteristics such as sex or region did not appear to significantly affect smartphone overdependence levels. Research implications or Originality - While there are some studies that predict smartphone overdependence level using machine learning techniques, but the studies only present algorithm performance based on survey data. In this study, based on the information gain measure, questions that have more influence on the smartphone overdependence level are presented, and the performance of algorithms according to the questions is compared. Through the results of this study, it is shown that smartphone overdependence level can be predicted with less information if questions about smartphone use are given appropriately.

Expression Analysis of the csp-like Genes from Corynebacterium glutamicum Encoding Homologs of the Escherichia coli Major Cold-Shock Protein CspA

  • Three csp-like genes were identified in the Corynebacterium glutamicum genome and designated cspA, cspB, and cspA2. The genes cspA and cspA2 encode proteins, comprising of 67 amino acid residues, respectively. They share 83% identity with each other. Identity of those proteins with Escherichia coli Csp proteins was near 50%. The cspB gene encodes a protein composed of 127 amino acids, which has 40% and 35% sequence identity with CspA and CspA2, respectively, especially at its N-terminal region. Analysis of the gene expression profiles was done using transcriptional cat fusion, which identified not only active expression of the three genes at the physiological growth temperature of $30^{\circ}C$ but also growth phase-dependent expression with the highest activity at late log phase. The promoters of cspA and cspA2 were more active than that of cspB. The expression of the two genes increased by 30% after a temperature downshift to $15^{\circ}C$, and such stimulation was more evident in the late growth phase. In addition, the cspA gene appeared to show DNA-binding activity in vivo, and the activity increased at lower temperatures. Interestingly, the presence of cspA in multicopy hindered the growth of the host C. glutamicum cells at $20^{\circ}C$, but not at $30^{\circ}C$. Altogether, these data suggest that cspA, cspB, and cspA2 perform functions related to cold shock as well as normal cellular physiology. Moreover, CspA and its ortholog CspA2 may perform additional functions as a transcriptional regulator.

Cloning and Characterization of Pseudomonas mucidolens Exoinulinase

  • An exoinulinase (${\beta}-D-fructofuranosidase$) gene was cloned by chromosome walking along the upstream region of the endoinulinase gene of Pseudomonas mucidolens isolated from soil. the exoinulinase gene consisted of an ORF of 0,506 bp encoding a polypeptide of 501 amino acids with a deduced molecular weight of 55,000. The exoinulinase produced by the recombinant Escherichia coli $DH5{\alpha}$ strain was also purified to homogeneity as determined by SDS-PAGE and a zymogram. The molecular weight of the purified exoinulinase according to both SDS-PAGE and gel filtration matched the deduced molecular weight of the protein described above, thereby indicating that the native form of the exoinulinase was a monomer. The purified enzyme hydrolyzed activity value of 2.0. Furthermore, no inulo-oligomers were liberated from the inulin substrate in the enzymatic reaction mixtures incubated for 90 min at $55^{\circ}C$. Taken together, these results indicate that the purified ${\beta}-D-fructofuranosidase$ was an exoinulinase. The pH and temperature optima of the exoinulinase were pH 6.0 and $55^{\circ}C$, respectively. the enzymehad no apparent requirement for a cofactor, and its activity was completely inactivated by $Ag^{+},{\;}Hg^{2+},{\;}and{\;}Zn^{2+}$. Kinetic experiments gave $K_m,{\;}V_{max},{\;}and{\;}K_{cat}$ values for inulin of 11.5 mM, 18 nM/s, and $72{\;}s^{-1}$, respectively. the exoinulinase was fairly stable in broad pH conditions (pH 5-9), and at pH 6.0 it showed a residual activity of about 70% after 4 h incubation at $55^{\circ}C$.

The Effects of Mulberry Fruit on the Antioxidative Defense Systems and Oxidative Stress in the Erythrocytes of Streptozotocin-Induced Diabetic Rats

  • The current study examined the effects of mulberry fruit on the antioxidative defense systems and oxidative stress in the erythrocytes of diabetes-induced rats. Sprague-Dawley male rats were randomly assigned to one normal and three streptozotocin (STZ)-induced diabetic groups. 1be diabetic groups were fed a mulberry fruit-free diet (DM-group), 0.3% mulberry fruit diet (DM-F group) or 0.6% mulberry fruit diet (DM-2F group). Diabetes was induced with STZ after three weeks of the experimental diets. 1be rats were sacrificed 9 days later for examination of the antioxidative defense systems and oxidative stress in the erythrocytes. Means of cy-3-Ο-glucopyranoside, cy-3-Ο-rutinoside, rutin, isoquercitrin, quercetin, morin and dehydroquercetin contents were 230.45, 131.5, 142.5, 10.3, 5.8, 1.6 and 3.83mg per l00g dry weight, respectively, in the mulberry fruit. Mulberry fruit strengthened the antioxidative defense systems through increased activity of the antioxidant enzymes, such as glutathione peroxidase (GSH-px) and catalase (CAT), in the erythrocytes of the diabetes-induced rats. Accrdingly, mulberry fruit was found to reduce the accumulation of thiobarbituric acid reactive substance (WARS). Therefore, mulberry fruit was found to be excellent for strengthening the antioxidative defense system and reducing damaging oxidative substances in the erythrocytes of the diabetes-induced rats.

Properties of the Arterial Pressor Response Induced by Stimulation of the Ventral Root Afferent Fibers in the Cat (고양이 척수 전근내 감각신경 자극으로 유발된 승압반응의 생리학적 특성)

  • In an attempt to characterize the ventral root afferent fibers, arterial blood pressure responses to stimulation of the ventral root (VR) were observed in anesthetized cats. Effects of the morphine administered either intravenously or direct spinally and of the spinal lesions on the pressor responses were compared. Followings are the results obtained. 1) Stimulation of the VR with C-strength, high frequency stimuli evoked a marked pressor response. No depressor response, which had been reported during peripheral nerve stimulation, was observed during VR stimulation with low frequency. 2) Acute cervical spinalization abolished the pressor response, indicating the involvement of supraspinal mechanism. 3) The ascending spinal pathways of the pressor response were located in the dorsolateral funiculus bilaterally. 4) Intravenously administered morphine exaggerated the pressor response to VR stimulation, while direct spinally administered morphine suppressed it. From the above results it was concluded that the ventral root afferent fibers have more similar properties to muscular C-afferent fibers than to cutaneous C-fibers.

Antioxidant Activity of Several Cabbage (Brassica oleracea L.) Cultivars

  • Total phenol, flavonoid and antioxidant components of cabbage leaf samples derived from different cultivar were determined. Total phenol compound content showed the highest amount in methanol extracts from ‘YR Howol’ cultivar (11.72 ㎎/g), followed by ‘Harutame’ (10.66㎎/g), ‘Winstar’ (10.34 ㎎/g) and YR Hero (10.20 ㎎/g). The highest amount of total flavonoid content was observed from the methanol extracts of Harutame (5.39 ㎎/g), followed by Winstar (4.28 ㎎/g), Wialhowol (4.10 ㎎/g). The SOD enzyme activity showed a high activity of ‘YR Hogeo’ cultivar, and the cultivar of ‘YR Howol’ cultivar showed the lowest activity of SOD. The activity of CAT and APX showed higher values ‘Ogane’ and ‘YR Hogeol’ cultivars than the other cultivars. The POD activities showed relatively high values ‘Ogane’ and ‘YR Howol’ cultivars compared with other cultivars. The free radical (DPPH) scavenging activity showed lower IC50 values of ‘Harutame’ (15.71) and ‘YR Howol’ cultivar (16.88), however methanol extract of ‘YR Hero’ cultivar (22.49) being the highest. The extracts of all cabbage cultivars in the reaction solution of pH 1.2 could be decomposed nitrite more than 50%. Especially, the cultivar ‘YR Hogeol’ and ‘Ogane’ showed a relatively high nitrite scavenging activity for each 60.13% and 57.20% respectively. The IC50 values of antioxidant activity determined by ABTS were lower in ‘Harutame’ (17.04) and ‘YR Howol’ cultivar (17.97), and its results observed similar with values obtained from the same extracts by DPPH method. The result of this study suggests that the methanol extract of Brassica oleracea L. contains the high amount of phenolic and higher radical scavenging activities.
