Objectives : This study aimed to evaluated the anti-inflammatory activity of the 70% ethanol extract from Vaccinium bracteatum leaves to investigate its effects on inflammatory mediators. Methods : Cell viability was assessed using an MTT assay in RAW 264.7 macrophages. Inflammatory responses were induced by lipopolysaccharide (LPS), and nitric oxide (NO) production was measured. The expression levels of inducible nictric oxdie synthase (iNOS) and cyclooxygenase-2 (COX-2) were analyzed by Western blotting, while interleukin-6 (IL-6) production was quantified using ELISA. Results : The ethanol extract exhibited no significant cytotoxicity up to 300 ㎍/mL. In LPS-stimulated RAW 264.7 cells, NO production was significantly inhibited in a concentration-dependent manner. In addition, the expression of iNOS and COX-2 proteins was markedly reduced, and IL-6 production was significantly suppressed following extract treatment. Conclusions : The ethanol extract of V. bracteatum leaves exhibited excellent anti-inflammatory activity by suppressing the production of NO and the expression of key inflammatory mediators, including iNOS, COX-2, and IL-6. These findings suggest its potential as a natural anti-inflammatory functional material.
