Evaluation of Antioxidant Activity and Tyrosinase Inhibitory Activity of Sapium macrocarpum Extract

Abstract

In this study, we evaluated the antioxidant activity and melanin production-related regulatory potential of Sapium macrocarpum methanol extracts (SMME), native to South America and Brazil. Antioxidant activity was determined by DPPH radical scavenging activity, reducing power, ·OH radical scavenging activity, and lipid peroxidation inhibition tests. SMME exhibited concentration-dependent antioxidant activity in all antioxidant tests, exhibiting DPPH scavenging activity of 36.1%, reducing power of 75.8%, and ·OH radical scavenging activity of 82.9% at 10.0 ㎍/mL. Cell viability tests showed no significant difference from the control group, confirming the absence of cytotoxicity within the effective concentration range. Western blot analysis showed that SMME reduced α-MSH-induced tyrosinase protein expression in a concentration-dependent manner, with a maximum decrease of approximately 94.7%. Furthermore, immunofluorescence staining demonstrated a decrease in tyrosinase fluorescence intensity, confirming the downregulation of tyrosinase at the protein level. Therefore, SMME is a candidate material derived from natural products with antioxidant activity and ability to control tyrosinase protein expression, and it can be used as basic data to explore mechanisms related to melanogenesis control.

본 연구에서는 남미 및 브라질 지역에 자생하는 Sapium macrocarpum 메탄올 추출물(Sapium macrocarpum methanol extracts, SMME)의 항산화 활성과 멜라닌 생성 조절 가능성을 평가하였다. 항산화 활성은 DPPH radical scavenging, reducing power, ·OH radical scavenging, lipid peroxidation 억제 실험으로 확인하였다. SMME는 모든 항산화 실험에서 농도 의존적인 활성 증가 경향을 보였으며, 10.0 ㎍/mL에서 DPPH 36.1%, 환원력 75.8%, ·OH 라디칼 82.9%의 활성을 나타냈다. 세포생존율 실험 결과, 유효 농도 범위 내에서 대조군과 유의한 차이가 없어 세포 독성을 유발하지 않았다. Western blot 분석에서 SMME는 α-MSH로 유도된 tyrosinase 단백질 발현을 농도 의존적으로 감소시켰고, 최대 농도에서 약 94.7% 감소하였다. 또한 면역형광염색 결과, tyrosinase 형광 강도의 감소가 관찰되어 단백질 수준에서의 발현 억제 효과가 확인되었다. 따라서 SMME는 항산화 활성과 tyrosinase 단백질 발현 조절 능력을 지닌 천연물 유래 후보 소재임을 시사하며, 멜라닌 생성 조절과 관련된 기전을 탐색하기 위한 기초 자료로 활용될 수 있을 것으로 판단된다.
