Abstract

Objective : Rutin is one of the most abundant compounds in the healthy vegetable broccoli. In this study, we isolated rutin from broccoli leaves, which are mostly discarded as waste, and investigated the neuroprotective effects of rutin on hydrogen peroxide (H2O2)-induced PC12 cells. Methods : Rutin was purified via bioassay-guided fractionation and confirmed by NMR. Apoptosis was determined by the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay, and the expression of related protein was monitored by Western blot analysis. Results : Pretreatment of PC12 cells with rutin resulted in significant protection against H2O2-induced cell death. Cell death induced by H2O2 was due to apoptosis as assessed by TUNEL assay. Rutin reduced TUNEL-positive apoptotic cells and restored the reduced expression of key apoptosis regulatory proteins, procaspase-9, -3, and PARP. In addition, rutin showed antioxidant effects by suppressing the increase in reactive oxygen species and the decrease in glutathione after exposure to H2O2. Moreover, rutin inhibited the activity of acetylcholinesterase, which causes memory and cognitive dysfunction. Conclusion : These results demonstrate that rutin has neuroprotective effects by modulating H2O2-induced apoptosis and oxidative stress, and also indicate the potential use of rutin as an antioxidant and neuroprotective agent.
