Abstract

Objectives : The purpose of this study was to investigate the antioxidant activity of water extract of Cistanches Herba (CH) in Leydig cells. Methods : We carried out the cytoprotective effect of CH in cultured mouse Leydig cells by MTT assay. Leydig cells treated with CH were incubated in the presence or absence of 100 μM hydrogen peroxide at 37℃ for 24 h. The protective effects of CH against hydrogen peroxide-induced oxidative stress, dihydrorhodamine 123 (DHR 123) assay, superoxide dismutase (SOD), catalase activity assays and lipid peroxide (LPO) were performed in Leydig TM3 cells. Results : Leydig cells treated with 5-250 ㎍/ml of CH showed no significant cytotoxicity. Leydig cells treated with 10 ㎍/ml of CH showed the most significant protective effect (49.0%, p < 0.05), decrease of intracellular levels of H2O2 (106.2%, p < 0.001) and decrease of LPO (1.87nmoles/mg protein, p < 0.05). Leydig cells treated with 10 ㎍/ml of CH showed increase of SOD activity and catalase activity but not significantly. Conclusions : These results suggest that CH has potent antioxidant activity on Leydig cells from H2O2-induced oxidative stress.
