Effect of varying ethanol concentrations on the extraction properties and physiological activity of Artemisia annua L.

에탄올 농도 조건이 개똥쑥 추출 및 생리활성에 미치는 영향

Abstract

The present study was undertaken to compare the effects exerted by different extraction solvents on the extraction of active components, such as polyphenols and flavonoids, from the dried leaves of Artemisia annua L. Different extracts were prepared using a heating mantle. The extraction solvents used were distilled water, and 20, 40, 60, 80, and 99.5% ethanol solution. It was observed that the 40% ethanol solution yielded the most significant results in the extraction of various phytochemicals with phenol concentration of 154.8±0.28 mg of gallic acid equivalent/g and flavonoid content of 25.28±0.01 mg quercetin equivalent/g. However, based on the extraction solvent used, varying trends were observed in the antioxidant, enzyme inhibition, and bacterial inhibition analyses. It was concluded that the extraction solvent should be selected based on the purpose of use of the dried leaves of A. annua L.

본 연구는 물과 다양한 에탄올 농도로 추출한 개똥쑥 잎 추출물의 페놀성 성분과 다양한 생리활성을 비교하여 효과적인 용매조건을 조사하기 위해 수행하였다. 총 phenol과 flavonoid 함량에서 40% 에탄올로 추출하였을 때 효율이 극대화되는 효과가 확인되었다. 이후 에탄올의 농도를 높이더라도 총 phenol 및 flavonoid 함량에 대한 긍정적 효과는 기대할 수 없었다. 인위적으로 생성시킨 DPPH, superoxide, nitric oxide 및 hydroxyl 라디칼소거능 실험 결과는 20, 40% 에탄올 추출물에서 높은 소거 활성이 나타났다. 반면 TEAC, FRAP 및 ORAC 분석을 통해 얻어진 총 항산화력은 60% 에탄올 추출물이 가장 높은 값을 보였다. 개똥쑥 잎 추출물의 항균활성은 물과 20% 에탄올 추출물에서는 생장저해효과가 없었지만, 40%에서 99.5% 에탄올 추출물은 활성이 관찰되었다. 효소 저해 활성의 경우 항당뇨와 관련이 있는 α-glucosidase 실험에서 99.5% 에탄올 추출물을 제외한 나머지 추출물은 양성대조구로 사용된 acarbose보다 비슷하거나 높은 저해능을 가진 것으로 확인되었다. 특히 60% 에탄올이 가장 높은 활성을 가졌다. Elastase 실험은 40%와 60% 에탄올에서 높은 활성을 보였고 tyrosinase는 용매로 사용된 에탄올 농도 의존적으로 저해능이 증가하는 경향이 있었다. 각 추출물에 대한 flavonoid 함량을 HPLC로 분석하였을 때 주요 flavonoid는 rutin, hesperidin, myricetin, nobiletin이었다. 또한 추출 용매에 따라 구성하고 있는 flavonoid의 성분이 달랐다. 결과를 종합하여 볼 때 개똥쑥 잎의 페놀성 성분은 극성-비극성의 넓은 범위를 가지며 중간 극성의 페놀성 물질의 함량이 높을 것으로 보인다. 그리고 개똥쑥 추출물의 생리활성은 강력한 활성을 가진 특정 flavonoid에 의해 나타났다. 이러한 연구 결과를 토대로 개똥쑥의 flavonoid 성분 추출 시 사용되는 용매 농도 설정에서 기초 자료로 이용이 가능할 것으로 판단된다.
