The Effects of Medicinal Herb-made Sikhe on Damage and Lipid Levels of Liver in Rats Fed High-cholesterol Diets

한약제 추출물로 제조된 식혜가 고지방식이에 의한 흰주의 간 손상과 간 지질 함량에 미치는 영향

Abstract

This study was conducted to evaluate the effect of Sikhe made by medicinal herb on the functional level of liver. Water extract I (12.9% W/W) and II (25.8% W/W) were obtained from medicinal materials: Caragana Sinica, Glycyrrhiza uralensis, Atractylodes rhizoma alba, Atractylodes rhizoma alba, Crataegus pinnatifida, Paeonia lactiflora Pasll., Hordeum vulgare Linne, Oryza sativa Linne, ginger, peer and jujube. Experimental groups were divided into the control diet group (C), high fat diet group (HF), high fat diet treated with 5% extract I group (HFE I ) and high fat diet treated with 5% extract II group (HFE II). In sensory evaluation, overall quality scores associated with color, aroma, flavor and taste were significantly higher in water extract II than in water extract 1. After investigating functional and lipid levels of livers in rats, we found that the administration of water extract I or water extract II to the high fat diet group (HF) did not affect the gain of body weight but mildly reduced GOT or GPT activity in the high diet group. Moreover, administration of these medicinal herbal extracts significantly decreased the levels of total lipid, triglyceride and total cholesterol in the high fat diet group (HF). However, administration of these medicinal herbal extracts did not affect the level of phospholipid. In conclusion, as Sikhe made by medicinal herb slightly decreased the activity of GOT or GPT and amount of lipid in liver, prevention against high fat diet is thought to be important for liver protection.

골담초, 작약, 백출, 감초, 맥아, 곡아, 산사의 한약재 추출물로 제조한 식혜의 관능검사와 이들 한약재 추출물이 고지방식이를 섭취한 흰쥐의 간기능과 간 조직내 지질함량에 미치는 영향에 대하여 조사하고자, 생후 7주령 숫쥐를 정상식이군(C군), 고지방식이군(HF군, 5% 대두유, 15% 쇠기름, 1% 콜레스테롤을 첨가한 식이) 및 고지방식이에 저농도와 고농도의 한약재 추출물을 각각 5% 첨가한 식이군(HFE I군, HFE II군)인 4군으로 나누었다. 한약재 추출물로 제조한 식혜의 관능검사와 실험식이로 8주간 사육한 결과는 다음과 같다. 식혜의 관능검사 결과는 저농도에 비해서 고농도의 한약재추출물로 제조한 경우 전체적인 풍미와 기호도 점수가 높은 점수를 보였다. 제조된 식혜의 영양소 함량은 시판되는 산업용 식혜에 비해서 열량, 탄수화물 그리고 단순당의 함량이 높게 나타났다. 동물실험에 있어서는 고지방식이에 의해 높아진 체중증가량에 대하여 한약재 추출물은 아무런 영향을 미치지 못하였다. GOT 활성도는 C군은 27.2${\pm}$3.83 karmen/ml, HF군은 66.7${\pm}$13.0 karmen/ml으로 고지방식이의 섭취 시 GPT 활성도는 p<0.05 수준에서 증가되었다. 그러나 HFE II 군의 GOT 활성도는 44.5${\pm}$16.1 karmen/ml로 HF군 (66.7${\pm}$13.0 karmen/ml)과 HFE I군(62.6${\pm}$5.61 karmen/ml)에 비해서 p<0.05 수준에서 낮은 활성도를 보였다. 또한 GPT 활성도는 C군에서 17.8${\pm}$4.61 karmen/ml로 가장 낮은 활성도를 보인 반면에, HF군에서는 52.8${\pm}$5.64 karmen/ml로 가장 높은 활성도를 보였다. 그러나 한약재 추출물 I과 II의 공급으로 GPT의 활성도를 다소 낮출 수 있었으나 정상 수준은 미치지 못하였다. 간 조직내 중성지질 함량에 있어서 C군은 63.7${\pm}$17.2 mg, HF 군은 136.9${\pm}$24.3 mg 로 고지방식이에 의해 중성지질 증가 현상이 나타났다 (p<0.05). 그러나 한약재 추출물 공급군인 HFE I 군과 HFE II 군의 중성 지질 함량이 각각 58.3${\pm}$13.8 mg과 51.1${\pm}$9.68 mg으로 C군과 비슷한 수준을 보였다. 간 조직내 총콜레스롤 함량에 있어서 C군의 22.8${\pm}$1.42 mg에 비해서 HF군은 40.8${\pm}$2.90 mg으로 고지방식이에 의해 간 조직내 콜레스테롤 함량은 p<0.05 수준에서 유의성 있게 증가되었다. 그러나 고지방 식이에 의한 고콜레스테롤 증가 현상은 한약재 추출물 I과 한약재 추출물 II의 공급으로 각각 30.1%와 44.6% 콜레스테롤 저하효과를 얻을 수 있었다. 인지질 함량은 C군은 17.6${\pm}$9.40 mg/g으로 가장 높았고 그 다음으로 HFE I군(17.4${\pm}$ 9.40 mg/g), HFE II군(14.5${\pm}$2.13 mg/g), HF 군(11.8${\pm}$7.45 mg/g) 순으로 낮아졌으나, 모든 실험군간에 유의적인 차이는 없었다. 이상의 결과를 종합해볼 때 식혜에 첨가된 한약재 추출물이 고지방식이에 의한 간 기능 지표 활성도와 간 지질 대사를 다소 개선시켰으나 정상수준에는 미치지 못한 것으로 나타났다.
