Abstract
Objective : Angelica Gigantis Radix has been clinically used to prevent or decrease the ischemia cerebral damage in Oriental medicine. The purposes of present study are to investigate whather Angelicae Gigantis Radix can protect against ischemia neural damage which was caused by middle cerebral artery occlusion(MCA0) in rats, and to investigate whether methanol extract of Angelicae Gigantis Radix has neuropmtective effect in primary cultured neurons in rats. Methods : By administration methanol of Angelicae Gaantis Radix, we compared treated groups and not treated groups in view of four points as follows : ① TTC staining and image analysis in rats subjected to pemanent focal cerebral ischemia induced by MCAO. ② LDH activities in the culture media on the primary cultured rat cortical neurons were treated with 30011M Glu or NMDA for l5min in the presence of the various concentratims of methanol extract of Angelicae Gigantis Radix. ③ LDH activities in the culture media on the primary cultured rat cortical neurons were treated with Fe2'/ascorbic acid, ha*, H a , xanthindxanthine oxidase, and SpermineNONOate in the presence of l00c(g/uit of the methanol extract of Angelicae Gigantis Radix. ④ LDH activities in that culture media on the primary cultured rat cortical neurons were treated with 30011 M Glu or NMDA for 15min in the presence of the various concentrations of methanol extract of decursin as 10, 30, 50, 100, 300㎍/dℓ. Result : The methanol extract of Angelicae gigantis Radix injected group showed significant induced in infartion areas and volume. And the primary cultured rat cortical neurons treated with Angelicae gigantis Radix extract were showed concentration-dependent of excitotoxic neuronal death. Conclusion : These findings indicate that Angelicae gigantis Radix can be used for treatment and prevention of ischemia.
