Isolation, identification, and culture conditions of the strain producing antibacterial antibiotic

Abstract

The strain with antibacterial activity was isolated among soil samples collected in Suwon area. The isolated strain was identified as Bacillus sp. YR-1 with respect to its morphological, cultural, and physiological characteristics. Optimal medium for the highest production of antibiotic was composed of sucrose 2.0%(w/v), peptone 2.0%(w/v) and NaCl 0.1%(w/v). The maximum production of antibiotic was shown at $35^{\circ}C$ for 48 hours with the initial pH 7.0.

식물에 세균성무름병(soft rot disease)과 검은반점병(blackleg)을 일으키는 Erwinia carotovora subsp. carotovora의 생육을 저해하는 균주를 토양으로부터 분리하고, 그 중 항균 활성이 가장 좋은 균주를 선별하여 균주의 동정, 항생물질의 최적 생산조건 및 생산된 항생물질의 특성을 연구 검토한 결과 다음과 같은 결론을 얻었다. 분리 균주의 형태학적, 배양학적 및 생리학적 특성을 검토한 결과 Bacillus sp.로 판단되어 분리균주를 Bacillus sp. YR-1로 명명하였다. 항생물질 생산을 위한 최적 조건을 검토한 결과, sucrose 2.0%(w/v), peptone 2.0%(w/v), NaCl 0.1%(w/v), pH 7.0의 배지 $35^{\circ}C$, 48시간 진탕배양 하였을 때, 항생물질의 생산성이 가장 좋았다.
