당뇨 흰쥐에서의 참마 $H_2O$ 분획물과 Selenium 보충시 혈당 및 지질대사에 미치는 영향

Effects or $H_2O$ Fraction or Dioscorea japonica Thunb with selenium on plasma Glucose and Lipid Metabolism in Streptozotocin Induced Diabetic Rats

초록

본 연구에서는 streptozotocin으로 당뇨를 유발시킨 횐쥐에게 methanol로 추출하여 분획한 참마의 $H_2O$ 분획물을 경구투여하고 selenium은 식이로 보충하여 당뇨시 14일간 혈당과 지질대사에 미치는 영향을 실험하여 다음과 같은 결과를 얻었다. 당뇨대조군은 당뇨로 인한 대사상의 변화로 정상군과는 다른 대사적인 차이를 나타내었다. 전 실험 기간 중 횐쥐의 체중변화는 정상군에 비해 당뇨실험군 모두에서 유의적으로 감소되었으며 DJT-Se군에서 다른 당뇨실험군보다 적은 양의 체중감소를 보였다. 참마와 Se 보충식이에 따른 식이효율도 DJT-Se군에서 유의적인 차이는 아니었지만 다른 당뇨실험군에 비해 높았다. 장기 의 무게를 체중 100g당으로 환산하였을 때 간장과 신장의 무게는 정상군보다 유의적으로 비대해졌으며 비장은 당뇨실험군 모두에서 정상군보다 무게가 낮게, 췌장은 높게 나타났다. 혈장 포도당 수준은 당뇨실험군 중 DJT-Se군에서 유의적으로 낮아졌으며 인슐린의 함량 수준은 당뇨실험군 간의 유의적인 차이를 보이지 않았으나 DJT-Se군에서 약간 높았다. 혈장단백질 수준은 당뇨대조군에 비해 당뇨실험군 모두에서 높게 나타났다. 혈장 지질 수준에 미치는 영향을 보면 혈장 cholesterol의 경우 당뇨대조군과 당뇨실험군 간의 큰 차이는 없었으나 당뇨실험군에서 낮은 경향을 보였다. HDL-cholesterol 수준은 모든 당뇨실험군에서 높게 나타났다. 중성지방의 경우는 DJT-Se군과 Se군에서 유의적으로 낮은 수준이었다. 유리지방산 함량은 정상군과 당뇨실험군 사이에 차이를 보이지 않았다. AST와 ALT 활성도는 군 간에 큰 차이를 보이지 않았으나 당뇨대조 군에 비해 DJT군에서는 유의성을 보였다. 본 연구 결과 참마나 Se를 독립적으로 섭취하는 것 보다는 병용하여 보충하는 것이 항산화 작용의 상승효과를 보여주어 고혈당과 지방대사산물에 작용하여 혈당강하효과를 보일 것으로 생각된다. 참마의 혈당강하에 영향하는 생리활성성분과 Se보충에 의한 대사적인 변화에 대하여 더 구체적인 생화학적인 연구가 이루어져야 될 것으로 여겨진다.

The purpose of this study was to investigate the effect of H$_2$O fraction of Dioscorea japonica Thunb(DJT) with selenium(Se) treatment on blood glucose and lipid metabolism in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats(180-220g) weighing were divided into five groups, that is one normal group and four diabetic experimental groups : the STZ-control group, the DJT group, the DJT-Se group and the Se group. Diabetes mellitus was induced in the male rats by injection of STZ into the tail vein at a dose of 45mg/kg B.W. The H,0 fraction of DJT(500mg/kg) given orally were administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na$_2$SeO$_3$(2mg/kg diet), which was prepared fresh daily. The body weight and food intake was monitored daily and plasma levels of glucose, insulin, hematocrit and protein were determined. The plasma concentrations of cholesterol, HDL-cholesterol, triglyceride and fire fatty acid were measured. The activities of aminotrans ferase were analysed. The body weight gain was shown to be significantly higher in the normal group than all diabetic groups. The blood glucose levels of the DIT-Se group was significantly lower compared to those of the experimental groups. The administration of H$_2$O fraction of DJT and selenium showed an increase in plasma protein concentrations. The plasma cholesterol levels of all STZ-groups were not significantly different and HDL-cholesterol levels were increased in the diabetic experimental groups fed on H$_2$O fraction of DJT or Se supplementation. Plasma triglyceride levels were lower in the DJT-Se and Se group than in the STZ-control group. free fatty acid levels were not significantly differ among STZ-control groups. STZ treatment increased aminotransferase activity and that of DJT group was highest. In conclusion, the data from the present experiments indicate that the treatment of the H$_2$O fraction of DJT with selenium showed a synergistic effect and the two can have an influence on hyperg1ycemia and lipid metabolites when administered together.
