Smart Polymeric Micelles as Nanocarriers for Gene and Drug Delivery

  • Polymeric micelles, supramolecular assemblies of block copolymers, are useful nanocarriers for the systemic delivery of drugs and genes. Recently, novel polymeric micelles with various functions such as the targetability and stimuli-sensitivity have been emerged as promising carriers that enhance the efficacy of drugs and genes with minimal side effects. This presentation focuses our recent approach to the preparation of functional block copolymers that are useful for constructing smart micellar delivery systems in advanced therapeutics, including chemo-gene therapy. Particular emphasis is placed on the characteristic behaviors of intracellular environment-sensitive micelles that selectively exert drug activity and gene expression in live cells.

  • Nanomedicine is a young and rapidly emerging field, which integrates clinical medicine with nanotechnology. Although the commercial nanomedicine is still in a fairly embryonic state, the recent advances in the nanotechnology-based therapeutics and diagnosis has changed the landscape of medicine. Bibliometric analysis shows a surge in research activity over the past decade. In this review, we have discussed some of the promising materials and their applications to this nascent field, such as carbon nanomaterials, polymeric drug delivery systems, and diagnostic imaging agents.

  • Polyvalent nanoparticle-DNA conjugates exhibit a variety of unique features such as programmable assembly and disassembly, sharp melting transitons, intense optical properties, high stability, enhanced binding properties, and easy fabrication of the surface nature by chemical and physical modification. The unique properties of nanoparticle-DNA conjugates enable one to build up a number of versatile assay schemes for the detection of various targets. In addition, nanoparticle-RNA conjugates also demonstrate great promise of therapeutic applications in the context of RNA interference when combined with polymeric materials. In this presentation, representative examples of each aspect of nanoparticle-oligonucleotide conjugates will be discussed.
