Preventive Effect of Sarcodon aspratus Extract on the Liver Damage in B(a)P-Treated Mice (벤조피렌을 투여한 마우스에서 향버섯 추출물의 간 손상 예방 효과)

  • To investigate the effect of Sarcodon aspratus methanol extract on liver damage in benzo(a)pyrene (B(a)P)-treated mice, mice were divided into 4 groups of control, B(a)P, Sarcodon aspratus methanol extract and Sarcodon aspratus methanol extract-B(a)P. The activities of serum aminotransferase, cytochrome P-450 and hepatic content of lipid peroxide after B(a)P-treatment were increased than control, but those levels were significantly decreased by the treatment of Sarcodon aspratus methanol extract. On the other hand, the hepatic glutathione content and glutathione S-transferase activity were increased by the treatment of Sarcodon aspratus methanol extract. Also the activities of superoxide dismutase, catalase and glutathione peroxidase were decreased by the treatment of Sarcodon aspratus methanol extract. In addition cytochrome P-450 1A1 isozyme protein level, which was remarkably increased by B(a)P treatment from results of immuno blotting, was decreased by the treatment with methanol extract of Sarcodon aspratus. These results suggest that Sarcodon aspratus methanol extract have protective effect on liver damage by decreasing lipid peroxide and activities of free radical generating enzymes.

The Effects of Regular Injection of High Dose Vitamin C on Liver Damage in Paraquat Poisoned Rat Model - Prehospital Care Model - (백서의 paraquat 중독모델에서 고용량 Vitamin C의 주기적 투여가 간에 미치는 영향 병원전 처치 모델)

  • Background: The toxicity of paraquat has been known to be caused by oxygen free radicals which leads to the lipid peroxidation and multiple organ failure. High dose vitamin C has been known to be a effective antioxidant activities against the paraquat intoxication. This study was designed to evaluate the effect of regular injection of high dose vitamin C on liver damage in paraquat poisoned rat model. Method: Fifty five rats of Sprague-Dawley strain were divided into three groups: control group, only intraperitoneally injected saline; paraquat group, single dose intraperitoneally injected paraquat(24.5%, 40 mg/kg) and every 8 hours injected saline; paraquat and vitamin C group, single dose intraperitoneally injected paraquat(24.5%, 40 mg/kg) and every 8 hours injected vitamin C(72 mg/kg). Rats were sacrificed on the 12 hours, 1st day, 2nd day, 4th day, 7th day after injection and liver tissue was obtained. H&E(Hematoxylin & Eosin) stain and Masson's trichrome stain for collagen fiber detection were undertaken. The results were observed using the microscope. Results: 1. There were no differences between control and experimental group at the 12hours after regular injection of high dose vitamin C. 2. There were significantly decreased liver damage in experimental group in the 1st day after regular injection of high dose vitamin C. 3. There were significantly increased recovery of liver damage with time in experimental group after regular injection of high dose vitamin C. Conclusion: These results suggest that regular injection of high dose vitamin C is effective in decreasing liver damage in paraquat intoxication.

Effects of Water Extracts from Lagocephalus wheeleri with Several Herbs on Hyperlipemia and Liver Damage Induced by Alcohol (은복과 한약재 복합물이 알콜 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향)

  • Objectives : This study was performed to investigate the effect of water extracts from Lagocephalus wheeled with several herbs(LW) on hyperlipemia and liver damage induced by alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And each extract was administrated for the same period. We measured the serum component in rat's blood, body weight and weight of liver. Results : At first, we observed effects of LW on hyperlipemia induced by alcohol. LW group didn't show significant increase of total cholesterol in comparison with those of the control group. LW group didn't show significant increase of HDL(High-Density lipoprotein) cholesterol in comparison with those of the control group. LW group showed significant decrease of triglyceride in comparison with those of the control group. LW group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. LW group showed significant increase of liver weight in comparison with those of the control group. At second, we observed effects of LW on liver damage induced by alcohol. LW group showed significant decrease of ALP, GOT, GPT and LDH in comparison with those of the control group. Conclusions : Reviewing these experimental results, it appears that water extracts from Lagocephalus wheeleri with several herbs have pharmaceutical efficacy on hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of this.

Effects of Saengkankeonbi-tang on Prevention of Hyperlipidemia and Liver Damage Induced by Alcohol (생간건비탕(生肝健脾湯)이 알코올 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향)

  • Objectives : This study was performed to investigate the effect of water extracts from Saengkankeonbi-tang on prevention of hyperlipidemia and liver damage induced by alcohol. Methods : Except for the normal group, we fed rats the control and Saengkankeonbi-tang groups with 25% alcohol for 55 days. For the same period we fed the Saengkankeonbi-tang group with Saengkankeonbi-tang extract as well and rats in normal and contrlo group on saline solution. We measured the serum components in rat's blood, body weight and weight of liver. Results : 1. At first, we observed effects of Saengkankeonbi-tang on prevention of hyperlipemia induced by alcohol. Saengkankeonbi-tang group showed significant decrease in total cholesterol levels in comparison with those of the control group. Saengkankeonbi-tang group showed significant increase in HDL-cholesterol levels in comparison with those of the control group. Saengkankeonbi-tang group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. 2. At second, we observed effects of Saengkankeonbi-tang on prevention of liver damage induced by alcohol. Saengkankeonbi-tang group showed significant decrease in GOT, GPT, LDH and ALP levels in comparison with those of the control group. Saengkankeonbi-tang group showed significant increase of liver weight in comparison with those of the control group. Conclusions : Reviewing these experimental results, it appears that water extracts from Saengkankeonbi-tang have pharmaceutical efficacy on prevention of hyperlipidemia and liver damage induced by alcohol.   • Prolonged alcohol consumption causes alcoholic liver damage due to the generation of reactive oxygen species, the accumulation of fatty acids, and an increase in inflammatory cytokines in the liver. In this study, the protective effect of a fruit extract of Paeonia anomala (FEPA) against chronic alcohol-induced liver damage was evaluated in Sprague-Dawley rats fed an ethanol or a control Lieber-DeCarli diet for 5 weeks to induce alcoholic liver damage. FEPA (50, 25, and 10 mg/kg body weight/day) as well as the reference control silymarin (25 mg/kg body weight/day) were administered along with the ethanol diet. FEPA protected against increases in alanine aminotransferase and aspartate aminotransferase in serum and attenuated alcohol-induced increases in triglycerides, tumor necrosis factor alpha, thiobarbituric acid-reactive substances, and cytochrome P450 2E1 enzyme activity in the liver compared with the group treated with ethanol only. Anti-oxidative defenses such as the total glutathione level and glutathione peroxidase activity were increased by FEPA treatment. These results suggest that FEPA exerts protective effects against chronic alcohol-induced liver damage by attenuating hepatosteatosis and pro-inflammatory cytokine production and enhancing anti-oxidative defense mechanisms in the liver.

Change of Dermal Oxygen Free Radical Metabolizing Enzyme Activities in Acute Liver Damage Induced with $CCl_4$ in Rats (급성 간손상의 실험동물 피부조직에 있어서 Oxygen Free Radical의 대사효소 활성 변동)

  • To evaluate an effect of liver damage on the dermal oxygen free radical metabolizing enzyme activities, the $CCl_4$ (0.1 ml/ 100 g body wt., 50% $CCl_4$ in olive oil) was intraperitoneally given to the rats every other day for 2 weeks. Based on the histopathological findings, liver weight (%), serum alanine aminotransferase, xanthine oxidase activities and hepatic lipid peroxide contents, the animals were induced severe liver damage. In the present liver damaged animal, all the activities of dermal scavenging enzymes such as superoxide dismutase, catalase and glutathione peroxidase were significantly decreased compared with central. And under the cytochemical electron microscopy the more coarse granules of cerrous perhydroxide were found compared with the control. In conclusion, the $CCl_4$-induced liver damage may influence upon the activities of some dermal oxygen free radical scavenging enzymes.

Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And each extract was administrated for the same period. We measured the serum component in rat's blood, body weight and weight of liver. Results : 1. At first, we observed effects of Injinho-tang and Haninjinho-tang on hyperlipidemia induced by alcohol. Injinho-tang group showed significant decrease of triglyceride, body weight in comparison with those of the control group. Haninjinho-tang group showed significant decrease of total cholesterol, triglyceride in comparison with those of the control group. I 2. At second, we observed effects of Injinho-tang and Haninjinho-tang on liver damage induced by alcohol. lnjinho-tang group showed significant decrease of AST (aspartate aminotransferase), ALT (alanine aminotransferase), ALP (alkaline phosphatase), liver weight in comparison with those of the control group. Haninjinho-tang group showed significant decrease of AST (aspartate aminotransferase), ALT (alanine aminotransferase) in comparison with those of the control group. Conclusion : Reviewing these experimental results, it appears that water extracts from Injinho-tang and Haninjinho-tang have pharmaceutical preventive efficacy on hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.

Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And the extract was administrated for the same period. We measured the serum components in rat's blood, body weight and weight of liver. Results : At first, we observed effects of PMR on prevention of hyperlipidemia induced by alcohol. PMR group showed significant decrease of total cholesterol and triglyceride in comparison with those of the control group.
