Effects of Fucoidan on Neuronal Cell Proliferation: Association with NO Production through the iNOS Pathway

  • Fucoidan, that is high-molecular-weight sulfated polysaccharides extracted from brown seaweeds has been shown to elicit various biological activities. Here, we investigated the effects of fucoidan on cell proliferation and nitric oxide (NO) production in neuronal blastoma cell (SH-SY5Y). In the present study, we demonstrated that fucoidan treatment resulted in increase of cell proliferation and NO production. When cells were treated with amyloid-${\beta}$ (A${\beta}$) in the absence or presence of fucoidan, fucoidan recovered the cell viability decreased by A${\beta}$ peptides. To further determine whether nitric oxide synthase (NOS) is involved in proliferative effect of fucoidan, cells were treated with NOS inhibitors in the absence or presence of fucoidan. Selective constitutive nitric oxide synthase (cNOS) inhibitor, diphenylene iodonium chloride (DPI), caused a decrease of cell viability, whereas cell viability was increased by specific inducible nitric oxide synthase (iNOS) inhibitor, S-methylisothiourea (SMT), in the fucoidan-untreated cells. Treatment with fucoidan inhibited the cell viability decreased in DPI-exposed cells. In contrast, fucoidan had no effect on cell growth in SMT-treated cells, indicating that cNOS may not play a role in the proliferation of fucoidan-treated cells. The present data suggest that fucoidan has proliferative and neuroprotective effects and these effects may be associated with iNOS.

Reduced Leptin Secretion by Fucoidan-Added Kochujang and Anti-adipogenic Effect of Fucoidan in Mouse 3T3-L1 Adipocytes

  • In order to improve the antiobesity effect of Kochujang, 1% of sea tangle powder, alginic acid extract, and fucoidan extract were added to Kochujang. Sea tangle powder-added Kochujang decreased leptin secretion by only 12% compared to Kochujang, whereas alginic acid or fucoidan-added Kochujang significantly decreased leptin secretion by more than 60% in 3T3-L1 adipocytes. Fucoidan, one of the active components of sea tangle, decreased leptin secretion by 56%, 60%, and 60% compared to the control in the concentrations of $1{\mu}M,\;2.5{\mu}M,\;and\;5{\mu}M$, respectively. To see the effect of fucoidan on TG formation during adipocyte differentiation, 3T3-L1 cells were treated with $1{\mu}M\;and\;5{\mu}M$ concentrations of fucoidan during adipocyte differentiation (from 'day 0' to 'day 6'). Oil red O staining showed fucoidan decreased the amount of TG droplets and $5{\mu}M$ fucoidan potently inhibited TG formation. To see the effect of fucoidan on lipolysis, differentiated 3T3-L1 adipocytes were treated with fucoidan. The secretion of glycerol, which is used to measure lipolytic activity, was increased by 21%, 37%, and 53% compared to the control in the concentrations of $1{\mu}M,\;2.5{\mu}M,\;and\;5{\mu}M$, respectively. Oil red O staining showed fucoidan decreased TG amount at $1{\mu}M\;and\;5{\mu}M$ concentrations. These results suggest that fucoidan decreases leptin secretion and TG accumulation by inhibition of adipocyte differentiation and induction of lipolysis. Since fucoidan is reported to have various biological activities in addition to an anti-adipogenic effect, it seems valuable to develop fucoidan-added Kochujang as a multi-functional Kochujang.

Fucoidan Enhances the Survival and Sustains the Number of Splenic Dendritic Cells in Mouse Endotoxemia

  • Fucoidan is a sulfated polysaccharide derived from brown algae that has been reported to perform multiple biological activities, including immunostimulation. In this study, we investigated whether fucoidan has beneficial effects on endotoxemia induced by LPS, a septic model in mice. The focus of this study was on survival rates and spleen function of the mice upon treatment. We found that fucoidan had prophylactic effects on the survival rate of mice with endotoxemia. Flow cytometric analysis using antibodies for subset-specific markers revealed that fucoidan profoundly reversed the depleted population of dendritic cells in mice with endotoxemia. According to Western blot analysis, the spleen cells of LPS/fucoidan-treated mice showed a higher expression of anti-apoptotic molecules compared to those of LPS-treated mice. Also, fucoidan-treated spleen cells were more responsive to mitogens. Taken together, these results demonstrate that fucoidan pre-treatment has beneficial effects on the survival rate and function of the spleen in mice with endotoxemia. This study may broaden the use of fucoidan in clinical fields, especially endotoxemia.

Increased Anticancer Activity by the Surfated Funcoidan from Korean Brown Seaweeds (한국산 길조류에서 추출한 Fucoidan의 황산기에 따른 항암작용)

  • Fucoidan is a kind of polysaccharides in brown seaweeds. For the past years have been extensively studied due to their numerous biological activities : anticancer, anticoagulant, antithrombotic, anti-inflammatory and antiviral. In this study, we h ave extracted fucoidan from the Korean brown seaweeds and examined it's anticancer activities for employed SV40 DNA replication assay, RPA-ssDNA binding assay of replication protein A(RPA: known as human single-stranded DNA-binding protein essential for DNA rep-lication) and MCF7 cell growth inhibition assay. In addition to, we found that chemically sulfated fucoidan'santicancer activity is more higher than natural and desulfated fucoidan. It seem that fucoidan's sulfate group affect on DNA replication, cause of decrease RPA's DNA binding activity. These results suggests that sulfated fucoidan from Korean brown seaweeds have anticancer activity.

Degradation of Fucoidan by Contact Glow Discharge Electrolysis Using Organic Electrolyte (유기산전해질을 이용한 접촉 글로우 방전 전기분해공정에서 후코이단의 저분자화)

  • Depolymerization of fucoidan induced by contact glow discharge electrolysis(CGDE) was investigated. To utilize fucoidan as a functional food material after CGDE, organic acids were used as a electrolyte in CGDE process. Experimental results showed that CGDE using organic acid reduced the molecular weight of fucoidan effectively. As electrolyte concentration increased, onset voltage of glow discharge decreased and onset current of glow discharge increased. From the variation of molecular weight of fucoidan with the reaction time, it was demonstrated that the degradation of fucoidan followed a first-order rate law. Molecular weight of fucoidan treated with CGDE using organic acid was about 77 times lower compared to initial fucoidan with little free sulfate.

Fucoidan Protects LLC-PK1 Cells against AAPH-induced Damage

  • This study was designed to investigate the protective effect of fucoidan against AAPH-induced oxidative stress in LLC-PK1 cells (porcine kidney epithelial cells). Oxidative stress was induced by exposing of LLC-PK1 cells to the 1 mM 2,2'-azobis(2-amidino propane) dihydrochloride (AAPH) for 24 hr. Exposure of LLC-PK1 cells to 1 mM AAPH for 24 hr resulted in a significant (p<0.05) decrease in cell viability, but fucoidan treatment protected LLC-PK1 cells from AAPH-induced cell damage in a dose dependant manner. To investigate the protective action of fucoidan against AAPH-induced damage of LLC-PK1 cells, we measured the effects of fucoidan on lipid peroxidation and antioxidant enzymes activities of AAPH treated cells as well as scavenging activities on superoxide anion radical and hydroxyl radical. Fucoidan had protective effect against the AAPH-induced LLC-PK1 cellular damage and decreased lipid peroxidation and increased activities of antioxidant enzymes such as superoxide dismutase (SOD) and glutathione peroxidase (GSH-px). Furthermore, fucoidan showed strong scavenging activity against superoxide anion radical. The $IC_{50}$ value of fucoidan was $48.37{\pm}1.54\;{\mu}g/mL$ for superoxide anion radical scavenging activity. The fucoidan also had high hydroxyl radical scavenging activity ($IC_{50}=32.03\;{\mu}g/mL$). These results indicate that fucoidan protects against AAPH-induced LLC-PK1 cell damage by inhibiting lipid peroxidation, increasing antioxidant enzyme activities and scavenging offree radicals.

Purification of Fucoidan from Korean Sea Tangle (Laminaria religosa) and Isolation of Fucoidan-Degrading Microorganisms (한국산 다시마 유래 Fucoidan의 정제 및 분해균의 분리)

  • The fucoidan from Laminaria relicollected at Wando in Korea was purified with the yield of 2.3% in mass. The monosaccharide composiof the purified fucoidan was nearly identical to that of the commercial standard: fucose 63.71 %, xylose 22.98%, galactose 6.62%, mannose 0.24%, and uronic acid 3.26%. Microorganisms capable of degrading the purified fucoidan were isolated from the colonies on the minimal medium containing 0.2% of purified fucoidan as a sole carbon source. Of these isolates, a strain showing a relatively higher capability to degrade fucoidan, up to 63%, was partially characterized as a Gram positive, aerobic, moderately halophilic marine bacterium.

Antitumor Effects of Fucoidan on Human Colon Cancer Cells via Activation of Akt Signaling

  • We identified a novel Akt signaling mechanism that mediates fucoidan-induced suppression of human colon cancer cell (HT29) proliferation and anticancer effects. Fucoidan treatment significantly inhibited growth, induced G1-phase-associated upregulation of p21WAF1 expression, and suppressed cyclin and cyclin-dependent kinase expression in HT29 colon cancer cells. Additionally, fucoidan treatment activated the Akt signaling pathway, which was inhibited by treatment with an Akt inhibitor. The inhibition of Akt activation reversed the fucoidan-induced decrease in cell proliferation, the induction of G1-phase-associated p21WAF1 expression, and the reduction in cell cycle regulatory protein expression. Intraperitoneal injection of fucoidan reduced tumor volume; this enhanced antitumor efficacy was associated with induction of apoptosis and decreased angiogenesis. These data suggest that the activation of Akt signaling is involved in the growth inhibition of colon cancer cells treated with fucoidan. Thus, fucoidan may serve as a potential therapeutic agent for colon cancer.

Properties of Fucoidan as Raw Materials of Water-holding Cream and Cosmetics (후코이단의 보습제 및 화장품재료로서 특성)

  • Properties of fucoidan used for functional cosmetics agents were studied. Fucoidan was extracted from Undaria pinnatifida sporophylls. To test possibility of fucoidan as a cosmetics material, water-holding property measurement, DPPH free radical scavenging assay and MTT assay were done. Water-holding property of fucoidan was higher than that of hyaruronic acid, which is known as the one of the best water-holding material. The water-holding strength of fucoidan slightly increase as molecular weight of fucoidan decrease. Fucoidan showed high stability from MTT assay and good anti-oxidation property from DPPH assay. To evaluate the effect of water-holding property and anti-alergy property of fucoidan on the atopic dermatitis(AD), 46 AD patients were treated with fucoidan cream. After 6 weeks treatment, Investigation Global Assessment(IGA) scores decreased from 3.04 to 2.15, that is fucoidan cream had a 39.8% benefit effect on atopic dermatitis.

Effect of Fucoidan Extracted from Hizikia fusiforme on Intestinal Villi and Salmenolla spp. in Broiler Chicks (톳추출 Fucoidan이 병아리 장내 Villi 및 Salmonella 균주에 미치는 영향)

  • This study was conducted to investigate the in vivo and in vivo antibiotic effect of crude fucoidan extracted from Hizikia fusiforme, and to investigate any possible structural changes of broiler chick's intestinal villi by the supplementation of fucoidan. Total 84 broiler chicks were randomly assigned to 7 treatments, control and Salmonella typhimurium infection groups. The broiler chicks was infected with Salmonella typhimurium at third days, and antibiotics, fucoidan, dried Hizikia fusiforme, dried Undaria pinnatifida and yeast cell debris was respectively supplemented for each group. Each treatment had 4 chicks with three replications. Extraction yield of crude fucoidan from Hizikia fusiforme was 5.453%. Antibiotic effect of fucoidan was not detected in vitro, inhibition zone and micoorganism growth test. Weight gains of broiler chicks were tend to higher in fucoidan treatment group and yeast cell significance was not found. In in vivo test, the number of viable Salmonella typhimurium was low in the antibiotics and fucoidan treatment groups. The intestinal villi were short in the fucoidan and marine algae treatment groups. The intestinal villi were densely distributed on the large intestinal wall, but the morphology was not different among treatments.
