Early Life History Characteristics of an Induced Hybrid between Rhodeus uyekii and Rhodeus ocellatus (각시붕어(Rhodeus uyekii)와 흰줄납줄개(Rhodeus ocellatus) 잡종의 초기생활사 특징)

  • This study was conducted to identify taxonomic differences in the characteristics of Rhodeus uyekii and Rhodeus ocellatus during their initial life history via an interspecific hybridization experiment. Hybrids were compared to their parent species, and the findings were used to inform developmental research in commercially useful aquarium fish. The hatching rates of the cross-bred eggs were 75.9% for cross UO (R. uyekii ♀${\times}$R. ocellatus♂) and 71.9% for cross OU (R. ocellatus♀${\times}$R. uyekii♂), which did not differ greatly from the hatching rates of the normal cross-bred group. Backcross experiments resulted in 100% egg mortality during development. Newly hatched larvae of the original hybrid crosses were similar to those of the maternal line, and the color of the egg yolk was similar to that of the paternal line; therefore, the respective traits of the interspecific parents were identifiable within the cross-bred offspring.

  • The osteological features of Rhodeus uyekii were examined. In this study, the detailed descriptions of cranium, vertebra and fin skeleton of Rhodeus uyekii are presented. R. uyekii had the following characteristics ; there was no fontanell in the cranium ; there was no process of supraethmoid ; the free interneural spine was large and flat ; the dorsal process of the supraoccipital was high and triangular ; the fourth infraorbital was reduced ; in the urohyal, the hypohyal attachment is bifurcated, the horizontal and vertical plate has a elongated rhombus shape with posterior edge pointed ; there was no coracoid foramen in the shoulder girdle ; there was no uroneural in the first preural centrum.

  • A intergeneric hybrid between Rhodeus uyekii and Acheilognathus signifer was collected in the Jojongcheon Bukhan River. The hybrid showed intermediate characteristics of two parental types in some key characters, such as color pattern of body, pigmentation of dorsal, anal, and caudal fins, and body spot and stripe. In addition, the hybrid had a fairly large number of lateral line scales, while it lacked a barbel, which is known as a key intergeneric character. The probable occurrence of a natural hybrid is supported by a viable $F_1$ hybrid produced with the parental species.

  • The purpose of this study is to investigate the early life history of the Rhodeus fish, Rhodeus uyekii and R. ocellatus, in the Nakdong River to use as the preliminary data for the systematic study. The embryos used in the study were fertilized eggs (embryo) and larvae after artificial fertilization. The long diameter of the eggs of the R. uyekii was 3.39-3.97 mm (average $3.68{\pm}0.41mm$, n=30) and the short diameter was 1.36-1.55 mm (average $1.45{\pm}0.13mm$, n=30). The long diameter of the eggs of the R. ocellatus was 2.53-2.71 mm (average $2.62{\pm}0.12mm$, n=30) and the short diameter was 1.47-1.60 mm (average $1.53{\pm}0.09mm$, n=30). Hatching time was 48 hours for the R. uyekii and 50 hours for the R. ocellatus given that the average water temperature was $21.5^{\circ}C$. The hatched larvae were 4.95-5.00 mm (average $4.98{\pm}0.04mm$, n=5) for the R. uyekii and the total length was 3.66-3.69 mm (average $3.67{\pm}0.02mm$, n=5) for the R. ocellatus. R. uyekii was found to be 15.5-15.8 mm at total length (average $15.6{\pm}0.21mm$, n=5) on the 56 days after hatching with the number of dorsal fins being ⅲ-9, anal fins ⅲ-10, ventral fins ⅲ-5. The R. ocellatus was found to be 15.8-16.0 mm (average $15.9{\pm}0.14mm$, n=5) at total length on the 58 days with the number of dorsal fins being ⅲ-11, anal fins ⅲ-12 and ventral fins ⅲ-5 where the number of all fin stalks reached maximum.

  • In this study we conducted artificial hybridization between two sibling species, Rhodeus uyekii and R. notatus, and observed the morphological characteristics in early developmental stage. The two species showed difference in egg shape having rhombus and club respectively. The morphology of yolk of larva just hatched also showed well specific characteristics, that is the bean chaff type for R. uyekii (U type) and anchor type for R. notatus (N type). The rate of fertilization between female R. uyekii and male R. notatus (UN type) and between female R. notatus and male R. uyekii (NU type) were complete and its hatching rate were very high, 71.6% for UN type and 97.5% for NU type. The differences occurred in the yolk shape of hatched larvae for each combination of hybrids. In the group of UN, U type of larvae were found very rarely, but almost all the larvae showed intermediate shape polarized to NN type. Similar phenomenon was observed in the NU having intermediate polarized to UU, but without any NN type. These hybrids will be analyzed for their external morphology, sex ratio, the function of sexual organ and karyology after they grown up to adult.

Elongation of the Ovipositor in Korean Rose Bitterling, Rhodeus uyekii (Pisces: Cyprinidae) (각시붕어, Rhodeus uyekii (Pisces: Cyprinidae)의 산란관의 신장)

  • Factors influencing elongation of the ovipositor in the bitterling Rhodeus uyekii were examined under several conditions. The ovipositors of the females were periodically elongated under the presence of mussels or males, and in some cases without either. When females were reared without mussels and males, the ovipositor did not elongate in one fish in a large aquarium, but it did become elongated in another fish in a small aquarium and in many fishes in a large aquarium. Duration of the elongated state of the ovipositor was 1~3 days (mean 1.2~2.5 days) and the periodicity of elongation was 4~15 days (mean 6.4~11.9 days). Length of the elongated ovipositor was 68~100% (mean 78.5~98.8%). Length of ovipositor in the interpeak period was 20~51% (mean 27.3~33.1%); in the post-elongation period it did not elongated further 3~30% (mean 11.1~19.9%). The effect of aquarium size and number of individuals in the aquarium on the elongation of the ovipositor, and inter-specific or inter-generic differences based on previous reports are discussed.

Effects of Dietary Carotenoids on the Nuptial Color of the Bitterling (Rhodeus uyekii) (Carotenoids 첨가 사료가 각시붕어 (Rhodeus uyekii)의 체색에 미치는 영향)

  • The body color of fish fed were improved to be clearer by feeding the bitterling an artificial diet containing different carotenoids. The possibilities of using the bitterling, Rhodeus uyekii, an aboriginal fish of Korea, were studied by feeding the experimental diets supplemented with the various carotenoids (astaxanthin, lutein, and $\beta$-carotene). Amount of total carotenoids in the groups of fish fed by supplementation carotenoids were relatively higher than the control group of fish fed by no supplementation carotenoids. Especially, the group of fish fed by lutein supplementation diet showed most clarity in body color. The group of fish fed by astaxanthin supplementation diet showed the highest growth while the group of fish fed by lutein or $\beta$-carotene supplementation diet showed no significant different comparing to the control group of fish. The results from this study indicated that the bitterling could be a new ornamental fish as long as provided with the proper diet and culture condition.

  • Eggs of Korean rose bitterling Rhodeus uyekii were collected and fertilized to observe temperature-related cleavage rates and mitotic intervals (${\tau}_0$). As the water temperature was increased, the slope of first cleavage frequency with elapsed time after fertilization increased, and approximately 30% of fertilized eggs reached first cleavage frequency at every 15 min. At higher temperatures, eggs developed faster and underwent further identical developmental processes. There were strong, negative correlations between ${\tau}_0$ and water temperatures at all temperatures studied (Y = -1.225X + 70.05, $r^2=0.988$, where Y is ${\tau}_0$ and X is temperature).

  • Reciprocal interspecific hybrids between two bitterling species Rhodeus uyekii (RU) and R. notatus (RN) were genetically identified based on the partial sequence analysis of recombination activating gene-1 (RAG-1) gene. Out of 863 bp positions analyzed, 13 nucleotide substitutions were detected between the two parental species (RU and RN genotypes). Both the induced hybrids (RU female$\times$RN male; UN genotype) and their reciprocal counterparts (RN female$\times$RU male; NU genotype) displayed the double peaks (or polymorphism) of sequence chromatograms at the 13 diagnostic positions, indicating that those hybrids were actual karyogamy derived from the two parental haploid genomes. However, it was not possible to distinguish between the reciprocal interspecific hybrids.

Fish Myogenic Regulatory Protein LUC7L: Characterization and Expression Analysis in Korean Rose Bitterling (Rhodeus uyekii)

  • Serine-arginine-rich nuclear protein LUC7L plays an important role in the regulation of myogenesis in mice. In the present study, we isolated and characterized the Korean rose bitterling Rhodeus uyekii Luc7l cDNA, designated RuLuc7l. The RuLuc7l cDNA is 1,688 bp long and encodes a 364-amino-acid polypeptide containing serine/arginine-rich region at the C-terminus. The deduced RuLuc7l protein has high amino acid identity (71-97%) with those of other species including human. Phylogenetic analysis revealed that RuLUC7L clustered with fish LUC7L proteins. The expression of RuLuc7l mRNA was high in the brain, kidney, and stomach of Korean rose bitterling. Expression of the RuLuc7l mRNA was detected from 1 day post-fertilization (dpf) and moderately increased until 21 dpf during the early development. Further investigations are required to elucidate the functional role of RuLUC7L in myogenesis in R. uyekii.
