  • Objectives : This study aimed to investigate the effects of Angelicae sinensis Radix on longitudinal bone growth rate in rats. We have screened traditional medicinal herbs to develop the longitudinal bone growth stimulator by well-established rat model. A. sinensis was identified as one of the effective herbs in the screening process. Methods : Adolescent female rats were administered A. sinensis at doses of 30 mg/kg and 300 mg/kg for 10 consecutive days. To observe the rate of longitudinal bone growth, tetracycline was injected intraperitoneally on day 8 to stain a fluorescent band on the anew formed bone. To elucidate the mode of action, we observed insulin-like growth factor-1 (IGF-1) and bone morphogenetic protein-2 (BMP-2) expression after A. sinensis administration in growth plate. Results : In the 300 mg/kg A. sinensis group, the length between the proximal endpoint of the tetracycline label and the division line between growth plate and bone was significantly increased compared with vehicle-treated control group. Height of the proximal tibial growth plate was higher in the A. sinensis group compared with control group. A. sinensis also upregulated the expressions of IGF-1 and BMP-2 in the proliferative zone and hypertrophic zone of the proximal tibial growth plate. Conclusions : A. sinensis increases longitudinal bone growth rate in rats. According to immunohistochemistry, A. sinensis increases local IGF-1 and BMP-2 expressions in the growth plate which can be considered as direct stimulation of GH on the local growth plate.

HT005 were consisted of the root of Angelica sinensis, the fruit of Alpinia oxyphylla, the root of Astragalus membranaceus, the stem of Eleutherococcus senticosus, and the root of Dioscorea japonica, and Poria cocos. Method : To investigate the effect on longitudinal bone growth in adolescent male Sprague-Dawley rats, the longitudinal length of growth plate was measured directly by the tetracycline fluorescent labeling method and chondrocyte staining method. HT005 administered orally for four days, and the tetracycline was injected twice on the growth plate of the animals for fluorescent dying. The rate of longitudinal bone growth was measured the length between the tetracycline bands which fixed on the 3rd day and 5th day after injection. Cresyl violet was also used to stain the chondrocytes in the growth plate. The length of growth plate after administration was compared. Expression of IGF-1 and BMP-2 in the growth plate was investigated by immunohistochemistry. Results : HT005 showed a significant longitudinal bone growth which was $301.0{\pm}6.1\;{\mu}m/day$ at the dose of 100 mg/kg and $283.8{\pm}1.25\;{\mu}m/day$ (p < 0.001)at the dose of 10 mg/kg of HT005, compared to control group by the tetracycline fluorescent labeling method. HT005 showed a significant chondrocyte length on growth plate which was measured $797.19{\pm}3.31\;{\mu}m$ (p < 0.001) at the dose of 100 mg/kg and $720.14{\pm}2.19$ (p < 0.001) ${\mu}m$ at the dose of 10 mg/kg compared to the control group by cresyl-violet staining method. Both the number and intensity of BMP-2 and IGF-1 positive cells were increased in the hypertrophic zone of growth plate. There was a significant correlation between BMP-2 and IGF-1 expression and heights of chondrocytes in growth plate. Conclusions : Treatment of HT005 on Sprague-Dawley rats markedly increased the longitudinal bone growth. Therefore, HT005 may be an alternative herbal source to growth hormone as it promotes bone growth in children afflicted by growth retardation.

  • The objective of this study was to examine the effect of polymer coating on the initial microorganism attachment and the biofilm growth. Such as nonion(polyacrylamine), anion(CMC-Na) and cation polymer coagulant(chitosan and PEI) were used for coating material of the support carrier(acryl plate). When polymer coagulant was coated with 5, 10, 20, 35, 50, 100 and 200 mg/l on the surface of acryl plate, initial microorganism attachment increased and optimum concentration for the attachment was 35 mg/l. Biofilm growth experiments were conducted with the substrate loading of 12.7gSCOD/$m^2\cdot$ day using RBC. The polymer coagulants such as CMC-Na, polyacrylamide, PEI and chitosan coating on the acryl plate facilitated the biofilm growth of microorganisms. Until the biofilm dry weight grows up to 0. 0038g/cm$^2$, biofilm growth on the plate coated with cation polymer like chitosan was better than that on the coated plate of nonion(polyacrylamine), anion(CMC-Na) polymer coagulant.

  • The purpose of this study was to suggest a systematic and scientific method for measurement of children's growth development, in which the accuracy of existing diagnosis method has not been concretely examined yet. The most popular method for diagnosis of children's growth is to analyze the opening degree of growth plate in each joint by X-ray image. However, X-ray method has some disadvantages; it is impossible to measure the diagnosis of growth periodically and repeatedly due to the radiation problem. Hence, this study introduced a profile analysis and the algorithm of analyzing the image of growth plate with the BUA(Broadband Ultrasound Attenuation) of calcaneus, to verify the possibility of alternative ultrasonic method harmless to human body. We obtained the images of growth plate in proximal tibiae, phalanges, and calcanei of 269 children (7∼16 years old) with X-ray. And the image of growth plate in calcanei was also obtained front those children with ultrasound. The results showed that the time of the opening degree of growth plate in each joint was almost consistent between X-ray and ultrasonic images. Also, the images of growth plate measured by X-ray and ultrasound showed the high correlation. Therefore, it is expected that the algorithm of ultrasonic profile analysis introduced in this study can replace the existing X-ray method to measure the growth plate correctly.
